Objective Heparanase (HPA) is an endo -β- D - glucuronidase that can degrade heparan sulfate proteoglycans (HSPGs) and has been implicated in tumor angiogenesis and metastasis. To investigate the effects of silencing heparanase on tumor growth, angiogenesis and metastasis of human gastric cancer in nude mice. Methods Human gastric cancer SGC - 7901 cells and human gastric cancer SGC - 7901 cells with silenced HPA (gastric cancer SGC -7901- HPA- ) were separately transplanted subcutaneously in 6 nude mice, then the growth time,growth size and growth speed of subcutaneous tumor were dectected. RT PCR and Western - blot were used separately to detect HPAmRNA and HPA protein expression of the two groups of subcutaneous tumors. Micro vessel density (MVD) of the two groups of subcutaneous tumors were detected by immunohist ochemical method . Two groups of subcutaneous transplanted tumors were separately injected into abdominal cavity of two groups of nude mice, with 6 mice each group. The growth of metastatic tumor in nude mice were oberserved. Results Subcutaneously transplantation of tumors were successfully built by human gastric cancer SGC-7901 cells and SGC-7901 -HPA- cells in nude mice ,and appeared separately at the 4th day and 7th day, the average volume of tumor at the 25th day were respectively (924±117. 6) mm3 and (486. 3±117. 5) mm3, MVD were respectively (20. 69±1.20) and (11.35±1.94) (P<0.05);HPAmRNA and HPA protein expression of subcutaneously transplanted tumor caused by human gastric cancer SGC- 7901 - HPA- cells were all decreased; Metastatic tumor of abdominal cavity caused by human gastric cancer SGC- 7901 cells were successfully built in three nude mice. Four metastatic lesions were found in abdominal cavity, including liver, right kidney, omentum and intestine, and the tumors were big. Metastatic tumor of abdominal cavity caused by human gastric cancer SGC - 7901 - HPA- cells were successfully built only in one nude mouse. Two metastatic lesions were found in abdominal cavity,including liver and right kidney,and the tumors were small. Conclusion Silencing HPA can inhibit the tumor growtruangiogenesis and metastasis of human gastric cancer in nude mice. It suggest that HPA may become a new approach for prevention and treatment of gastric cancer.
