[Objective]Gastric carcinoma remains one of the most common tumors in china. Surgical resection combined with chemotherapy or radiation therapy is the traditional therapeutic modality capable of cure, but the effect is not so good as we imagine. To change this grim outlook including to find new type of chemotherapeutics and to attempt various combination regimens is the developing trend of oncotherapy in future .Chinese herbal medicine has been applied in the clinical work of oncotherapy for a few years and has been proved that it is useful to inhibit the growth of tumor .In recent years more research is designed to determine the mechanism of Chinese herbal medicine in cancer treatment and prevention, and practitioners find some evidence from clinical trials that some Chinese herbs may contribute to longer survival rates, reduction of side effects, and lower risk of recurrence for some types of cancer, especially when combined with western anticancer treatments. Fructus amomi rotundus is one of the traditional Chinese medicines, which focuses on restoring a balance of body and aims to restore or strengthen immunity and resistance to tumor. According to the latest results, the extraction of fructus amomi rotundus can be used to damage cancer cell by destroying the protection factor of tumor and inhibit the propagation of SGC-7901 cell which comes from human gastric adenocarcinoma. This experiment is designed to explore the effect and mechanism of fructus amomi rotundus combined with 5-FU to the growth of gastric adenocarcinoma cell by building up orthotopic transplantation tumor of human gastric cancer in nude mice model , on basis of which to supply experimental evidence to prove that fructus amomi rotundus could be used as a type of anti-gastric cancer medicine.[Method]This study aimed to establish an orthotropic nude mouse model for human gastric cancer. Human gastric cancer cell lines SGC-7901 were raised in vitro and then were injected subcutaneously into 24 nude mice which were randomly divided into 4 groups when it grown into pieces in the nude mice (six in each): control group; 5-fluorouracil group(5-FU group); extraction of fructus amomi rotundus group(FAR group); combination group(5-FU+FAR group). Control group was given phosphate buffer solution(PBS) by the way of intragastric administration(dose 0.2 ml, once daily) and intraperitoneal injection (dose 0.2 ml, once every two days).FAR group was given fructus amomi rotundus by the way of intragastric administration (dose1.0g/(kg·d), once daily) and intraperitoneal injection (dose 20mg /(kg·d), once every two days).5-FU group was given both phosphate buffer solution(PBS) by the way of intragastric administration(dose 0.2 ml, once daily) and 5-FU by the way of intraperitoneal injection 20mg /(kg﹒d) once every two days).Combination group were given both fructus amomi rotundus by the way of intragastric administration (dose1.0g/(kg·d), once daily) and 5-FU by the way of intraperitoneal injection 20mg /(kg﹒d) once every two days).General condition such as volume and weight of the tumor were measured and recorded to dram a weight-time curve diagram .The nude mice were killed at the end of experiment. Volume and weight of the tumor were determined to calculate the inhibiting rate of tumor. Morphologic characteristics of tumor cell was observed by microscope under dyeing HE. The expression of Cyclooxygenase-2(COX-2) and Vascular Endothelial Growth Factor (VEGF) was detected by the way of immunohistochemisty. And CD34 was also detected to reflect the microvessel density(MVD).[Results]1. The model of orthotropic transplantation tumor of human gastric cancer is successfully built up in nude mice.2. The weight loss in 5-FU group, FAR group and Combination group is significantly different compared with that in control group, respectively(P<0.05).3. Volume of Tumor in Control group, FAR group, 5-FU group and Combination group is(1424.38±188.62)mm3, (800.52±163.55)mm3,(574.21±115.73)mm3 and(322.65±62.39)mm3 and weight of tumor in Control group, FAR group, 5-FU group and Combination group is (1.822±0.24)g,(0.987±0.20)g and (0.414±0.09)g, respectively。According to the results above , the mean volume and weight of tumor in FAR group, 5-FU group and Combination group is significantly different from that in control group(P<0.05). However the mean volume and weight in Combination group is significantly different from that in FAR group or 5-FU group(P<0.05) .4. The inhibition rate of 5-FU group, FAR group and Combination group is 45.83%, 63.67% and 77.28%.5. Nucleus of tumor cell in control group is obviously different with the characteristics of karyomegaly , anachromasis , heteromorphism and karyokinesis and it is less change in FAR group , 5-FU group and Combination group .6. VEGF, COX-2 and MVD in FAR group and Combination group were remarkablely different from that in Control group (P<0.05), but it was more significantly different between Combination group and Control group in statistics(P<0.01). [Conclusions]1. Inhibition on the growth of tumor is evidenced by not only single use of FAR or 5-FU but also combination of FAR with 5-FU .2. Synergistic effect is achieved by combination of FAR with 5-FU, it shows more remarkable inhibition on the tumor growth than either single agent.3. The possible mechanism of FAR to inhibit the growth of orthotopic transplantation tumor of human gastric cancer in nude mice may relate to reduce the expression of VEGF and COX-2 which are important to induce the formation of microvessel.
