ObjectiveTo investigate the effects of a Chinese compound prescription(i.e.,XianyuYizhi decoction) on the behavior and lipid peroxidation in hippocampusin epileptic rats and detect the safty of it by acute toxicology in Kunming mice.Methods1. The chronic epilepsy model was established by the intraperitonealinjection of pentylenetetrazole (PTZ). The rats were divided into four groups:saline group(NS), XianyuYizhi decoction group(XYYZ), Sodium ValproateOral Solution group(VPA),Sodium Valproate Oral Solution and XianyuYizhidecoction group(VPA+XYYZ)(n=15). Simultaneously, the establishment ofchronic epilepsy model were administrated by gastric perfusion daily for fiveweeks saline or drugs in each group. And then, the rats completely kindled wereinvestigated by Morris water maze test(MWMT). The level of malondialdehyde (MDA),glutathione (GSH) and Superoxide Dismutase (SOD) contents wereanalyzed by colormetric method.2. XianyuYizhi decoction was condensed into the largest concentration(5.75g/ml). And then by intragastric administration in mice, the median lethal(LD50)dose is not measured.So the maximum dosage of XianyuYizhi decoctioncan only be measured.Results1. The kindled and mortality rate of XYYZ group, VPA group,VPA+XYYZ group decreased compared with NS group,and the kindle latencyperiod is also significantly prolonged. Moreover, in MWMT the times of crossplatform of XYYZ group and VPA+XYYZ group were more than NS group.The platform quadrant staying time of drug administration groups were morethan NS group. MDA content in drug delivered group were significantlydecreased while the level of GSH and SOD activity were significantly increasedcompared with NS group (P<0.05).2. The responses of acute toxicological in mice did not appear. Themaximum dosage of XianyuYizhi decoction is384g/kg, equivalent to100timesof the clinical dose.ConclusionThe neuroprotective effect of XianyuYizhi decoction is by antioxidantinjury mechanism, to reduce the occurrence of epilepsy and improve cognitivefunction. XianyuYizhi decoction has very good security.
