OBJECTIVE: To study antioxidation and antiglycation of polysaccharides fromMisgurnus anguillicaudatus. METHODS: Polysaccharides from body of Misgurnusanguillicaudatus (MAPs) were extracted and separated into four parts which werenamed MAPⅠ, MAPⅡ, MAP Ⅲ and MAP Ⅳ, respectively. Polysaccharide fromsecretion of Misgurnus anguillicaudatus was named MAP Ⅴ. Purification, generalproperties and molecular weight of MAPs were measured, the scavenging ability ofMAPs to OH-· and O2-· were studied in the system of H2O2/Fe2+and pyrogallolauto-oxidation, respectively. Three stages of glycation reaction in glucose/BSAsystem were researched through NBT reductive ability, determination of dicarbonylcompounds and fluorescence of AGEs assay. SDS-PAGE assay was used to test theformation of glycated protein. RESULTS: The results showed that no absorption at260and280nm were observed in UV spectrum, and then typical absorption peaks ofpolysaccharides were determined in IR spectrum. There is obvious absorption peak ofpolysaccharides at490nm in the method of pHenol-sulfuric acid. All the MAPs hadinhibitory effect on oxidation and glycation and exhibited dose-dependent manner.Especially MAP Ⅳ displayed stronger antioxidation effect than glucosamine underexperimental conditions. Scavenging ability of MAP Ⅳ on OH-· was26.1%, and IC50of MAP Ⅳ on O2-· was0.681mg/mL. MAPs can inhibit the glycation from threeperiods. Especially, MAP Ⅳ showed an obviously inhibitory effect on the formationof dicarbonyl compounds. IR of MAP Ⅳ was72.5%, stronger than that ofaminoguanidine (p<0.05n=5). CONCLUSION: The functions of MAPs wereassociated with the content of glucosamine and molecular of MAPs. Oxidation and glycation are relevant with diabetic complications, the result of this work suggestedthat MAPs may be effective preventing and treating diabetic complications.According to the mechanism of diabetes complications, MAPs have the possibility tobecome inexpensive, relatively non-toxic therapies for the prevention and treatment ofdiabetic complications.
