Objective1.The main hypoglycemic constituents of Phellinus igniarius were the polysaccharides of Phellinus igniarius.In this study,the best combination of the extraction time,liquid ratio and extraction temperature was investigated and confirmed by single factor test and orthogonal test when the yield was the highest.2.The changes of DPPH free radical scavenging rate of crude polysaccharides extracted from Phellinus igniarius with the optimal extraction combination were studied and the effects of optimal combination on the activity of polysaccharide were studied.3.In vivo experiment was conducted to study the improvement effect of polysaccharides of Phellinus igniarius on type I diabetes mellitus male mice induced by streptozotocin(STZ)in different dose concentrations,in which the crude polysaccharide of Phellinus igniarius was extracted by the optimal extraction combination.Methods1.The polysaccharide from Phellinus igniarius is soluble in water with relatively high temperature,so study take the traditional water extraction method as the extraction of crude polysaccharides of Phellinus igniarius,to explore the effect that the extraction time,extraction temperature and liquid ratio have on the yield of the polysaccharide of Phellinus igniarius by single factor test and orthogonal test so as to determine the best factor-level combination with the highest yield of the crude polysaccharides of Phellinus igniarius.2.The polysaccharide extracts of Phellinus igniarius were extracted by different orthogonal test combinations to determine the optimum extraction combination of polysaccharide from Phellinus igniarius by spectrophotometry.3.The optimized extraction process was used to extract polysaccharides from Phellinus igniarius and then coarse polysaccharides was obtained by vacuum drying finally DPPH radical scavenging test was carried out at different concentrations of crude polysaccharide to determine the activity.4.In vivo hypoglycemic experiment was conducted on 25 male Kunming mice which were divided into five groups:blank group,group of model I type diabetes mellitus induced by streptozotocin and groups of high,medium and low dose of polysaccharide of Phellinus igniarius.Indicators of weight changes,levels of fasting blood glucose and levels of glucose tolerance were measured to study the effects of different doses of polysaccharide of Phellinus igniarius on hypoglycemic effect.ResultsThe optimal extraction conditions of crude polysaccharides from Phellinus igniarius were as follows:the ratio of material to liquid was 1:18,and the temperature of extraction water was at 90? and the extraction time was 8 hours.The results of antioxidant experiments showed that the crude polysaccharide of Phellinus igniarius had strong antioxidant capacity,and the highest clearance rate was 94.9%.The results of hypoglycemic experiment showed that the symptoms of diabetes mellitus had improved in streptozotocin-induced male Kunming mice of type I diabetes mellitus group,the blood glucose and drinking water have decreased,the body weight gain have recovered.The glucose tolerance of low dose group,middle dose group and high dose group was improved compared with diabetic group after three weeks of different doses of polysaccharide of Phellinus igniarius.ConclusionThe highest yield of extraction combination of the ratio of material to liquid,the temperature of extraction water and the extraction time was:the ratio was 1:18,the temperature of extraction water was at 90? and the extraction time was 8 hours.The DPPH free radical scavenging efficiency of polysaccharides of Phellinus igniarius showed a strong antioxidant capacity and the results of animal experiment showed that polysaccharides of Phellinus igniarius had a hypoglycemic effect on male type I diabetes mellitus Kunming mice induced by streptozotocin,and have enhanced the glucose tolerance of mice compared with the negative control group.
