| Objectives: The purpose of this paper is to study the effect and mechanism ofsinomenine(SIN), which is a selective COX2inhibitor, on proliferation and cell cyclearrest and apoptosis of non-small cell lung cancer(NSCLC) A549cell line and its synergyeffection with cisplatin. The paper intends to explore the function mechanism of SINantitumor in order to provide the theoretical basis for the prevention and treatment oflung cancer.Methods: Cultivate NSCLC cell lines A549cells in vitro, using MTT method todetect the cell growth inhibition rate of SIN (2.5,1.25,0.625mmol/L),cisplatin (0.625ug/ml) and combination treating NSCLC A549cell lines by24,48and72hours. Theflow cytometry(FCM) dyed by PI detects the cell cycle of SIN (2.5,1.25,0.625mmol/L)group and of its combination with cisplatin in processing A549cells after48hours.Annexin V/PI double dyeing method detects the cell apoptosis rate of SIN (2.5,1.25,0.625mmol/L) group and its combination with cisplatin in processing A549cells after48hours.Results:1. The proliferation effect of single drug groups of SIN and cisplatin andcombination on huaman lung adenocarcinoma A549cells: with the extension of time andthe increase of concentration, the proliferation inhibition of SIN and cisplatin andcombination on huaman lung adenocarcinoma A549cells respctively improve,displaying the dependence on time and dose. The effect of combination group is stronger than single medicine group, proving the two medicines have synergy effect. The longerthe time, the more obvious the synergy effect.2. Cell cycle experiments detecting SIN, cisplatin and combination on lungadenocarcinoma A549cells show that, compared with controls, the ratio of Gl periodcells gradually increases and ratio of S period cells gradually reduces, processed by SINof different concentration after48hours and with the increase of the drug concentration.the ratio of Gl period cells reduce and ratio of S period cells increases, processed bycisplatin.The ratio of Gl and S periods cells of combination are increase, providing astatistical significance.3. Annexin V-FITC/PI double dye detection cell apoptotic show that, comparedwith control, SIN(0.625,1.25,2.5mmol/L)and combination with cisplatin process A549cells after48hours, the early and late apoptotic cell numbers increased, and in a range ofconcentration, the proportion of apoptotic cells increased with the increase of SINconcentration.The percentage of apoptotic cells in combination is higher than in SIN.Conclusion: Sinomenine has the influence of proliferation inhibition on NSCLCA549cells and synergy after its combination with cisplatin. In a range of concentration,displaying the dependence on time and dose. Its mechanism may block the cell cycle inG1ã€S periods and induce apoptosis. |
