Objective:1.To oberve the different stages of Candida albicans biofilm morphology.2. To study the drug resistance of Candida albicans during differentstages of its biofilm formation.Methods:1.To establish the model of Candida albicans biofilms in vitro on thecoverslip, then investigate the biofilm forming process throughfluorescence microscopy and scanning electron microscopy.2.Using National committee for clinical laboratory standards(NCCLS)broth microdilution method to detect drug resistance of Candida albicansin the free state,4hBF and48hBF state.Results:1.Candida albicans can form typical biofilm on the coverslip.Byfluorescence microscopy and scanning electron microscope, we can see Candida albicans biofilms its cells and extracellular matrix graduallyincreasing with time extending, it can formed a typical biofilmarchitecture after48h cultured.2.The antifungal resistance of Candida albicans increasing with thebiofilm growing maturity.The order of fluconazole MICs is:48hBF>4hBF> free state, and the differences between the three were statisticalsignificant respectively.Conclusions:1.We can build the model of Candida albicans biofilm in vitro on thecoverslip and can be detected by fluorescence microscopy and scanningelectron microscopy.2.Antifungal resistance of Candida albicans biofilm is increasing with thebiofilm maturation.
