Abstract Objective:Chronic mountain sickness (CMS) is clinical syndrome which ischaracterized by polycythemia (Hb of male≥210g/L,Hb of female≥190g/L) andsevere hypoxemia, as settlers who live at altitude of2500meters gradually loseacclimatization to high altitude hypoxia. CMS pathogenesis has not been fully elucidated.Imbalance between generation and death of red blood cells may be the reasons of CMS. Studiesconfirmed that over-proliferation of red blood cells play role in CMS. Apoptosis studysof bone marrow hematopoietic cells carried out in recent years, Founding that apoptoticindex of bone marrow mononuclear cell was significantly decreased, and caspase-3caspase-8andcaspase-9expression decreased Survivin expression increased, suggesting that apoptosis ofhematopoietic cell is down-regulation in CMS.This study aims to understand physiological significance of Bax and Bakexpression levels in bone marrow, laying the foundation for the elucidation of theregulatory mechanism of hematopoietic cells apoptosis.Method:The samples of bone marrow used in this study were obtained from15male patientswith CMS(CMS group) and15healthy adults male (Control group).The expression of Bak andBax in bone marrows were detected using the SABC immunohistochemistry technique,to checkHb concentration in blood. Compare the expression of Bak and Bax in bone marrow of CMSgroup with Control group, to compare the proportion of positive cell between two groups. Toanalysis the liver correlation of expression level of Bak and Bax in bone marrows with Hbconcentration in blood.Results:Compared with the control group, accumulate points of the expressions of Bak and Baxwith the CMS group to have reduce, with a statistical significance (P<0.05). The proportion ofpositive cell in of Bak and Bax in bone marrows showed nothing of statistics significance. Therewere inverse correlations in the positive rates of Bak and Bax proteins in bone marrow, compareHb in blood in CMS group (P<0.05).Under microscope Bak and Bax proteins in bone marrowdistributed in cell and the intercellular substance. They major location in cytoplasm, scarce in cellmembrane. They show light brown and light yellow, assume piles of and diffuse matter, somethingstrong, and something weak. The cell of Bak with positive show brown granulates. They primarilyexist in cytoplasm, show more diffused.Conclusion: Compared with the control group, accumulate points of the expressions of Bak andBax proteins to have reduce in bone marrow in the patients with(CMS). There were inversecorrelations in the positive rates of Bak and Bax proteins in bone marrow, compare Hb in blood inCMS group (P<0.05). Result show abnormal of the expressions of Bcl-2family proteins. Theabnormal expression of Bcl-2family proteins in the pathological regulation of apoptosis inhematopoietic cells may play a role in the CMS in polycythemaia mechanism.
