Objective To observe the effects of EP to adjusting the glucolipid metabolism of Type2Diabetic Rats and discuss its possible mechanisms.Methods Rats model of type2diabetes was established by a high sugar and fat diet with intraperitoneal injection of STZ(30mg/kg.bw),and then given various doses of EP for28days.Recorded the intake of water and food every day;Measured the daily weight,the weekly FBG and OGTT;At the end of the experiment,evaluated the levels of INS,ISI,TC,TG,HDL,LDL,SOD,MDA and GSH-Px; Ob served the liver and pancreas organization structure by HE staining; detected the expression of GCKmRNA,InsRmRNA in liver tissue,and GLUT-4mRNA,APNmRNA in fat tissue by RT-PCR.Results1. General situationCompared with modle group,EP intervention groups’symptoms were lighter, including listlessness,Hyporichosis,polydypsia,polyphagia,polyuria,emaciation,etc. The effect of high and medium dose groups is especially remarkable.2. blood sugar indexCompared with control group,FBG and INS of model group were increased significantly,ISI declined obviously(P<0.01);Compared with model group,EP intervention groups’FBG were declined significantly,ISI were higher markedly (P<0.01);For the INS, high dose group was the only one different from model group (P<0.01);In the OGTT, compared with modle group,EP intervention groups’blood sugar could recover more quickly in2hours.3. blood fat indexCompared with control group,the level of TC, TG, HDL and LDL-C of EP intervention groups’were increased significantly (P<0.01);compared with model group,EP intervention groups’TG,high and medium dose groups’TC and LDL-C were reduced significantly (P<0.01).4. antioxidant indexCompared with control Group,model group’s MDA was higher (P<0.05),GSH-Px was lower significantly (P<0.01),SOD wasnot different (P>0.05);compared with model group,EP intervention groups’MDA were lower (P<0.01),SOD were higher (P<0.05), GSH-Px of high and medium dose groups were higher (P<0.01; P<0.05).5. HE staining of liver and pancreasHE staining of liver:model group showed the fuzzy structure of hepatic lobule and cords,most swelling and part of atrophy liver cells,a lot of sizes focally vacuolated lipid drops;compared with model group,high and medium dose groups’ liver cable were in order,liver cells were normal without fatty degeneration;low dose group had smaller amounts swollen,atrophic and adipohepatic liver cells.HE staining of pancreas:model group showed islet number was less,the volume was small,most islet cells have been atrophy and appeared fat empty bubble;compared with model group,EP intervention groups had more complete islet structure,more islet numberless pyknotic nucleus, especially for the high and medium does groups.6. expression level of mRNACompared with model group,the expression of InsR in EP intervention groups were lower (P<0.01);GCK expression in high and medium dose groups were higher (P<0.01);APN expression in medium dose group was higher than the model group(P<0.05);GLUT-4expression in EP intervention groups was no difference with model group(P>0.05),but the expression level had a rise trend with the increasing concentration.Conclusion:1.Continuous high sugar and fat fed and small doses of STZ intraperitoneal injection can almost copy type2diabetic rats model;2. EP can play a certain role in adjusting glycolipid metabolism in type2diabetic rats, relieve rats’diabetesrats syndromes-polydypsia,polyphagia,polyuria,emaciation, reduce blood sugar and blood fat,increase insulin levels,enhance insulin sensitivity, repair the liver and pancreas, etc;3. The possible mechanism to adjust type2diabetic rats’glucolipid metabolism of EP:(1) Enhance antioxidant effect, restrain oxidatie stress,repair the damaged islet and1steatosis liver;(2) Up-regulated APN and InsR expression in adipose and liver tissue to increases insulin sensitivity,improve the IR;(3) Up-regulated GLUT-4and GCK expression in adipose and liver tissue to promote the transport and metabolism of glucose.
