OBJECTIVE To investigate the expression and the relativity of RECK,RAGE and MMP-9 in immortalized human nasopharyngeal epithelial cell NP69-SV40T, and nasopharyngeal carcinoma (NPC) cells 5-8F(high oncogenicity, high metastasis) and 6-10B(oncogenicity, non-metastasis).METHOD RECK,RAGE and MMP-9 mRNA were detected with fluorescence quantitative polymerase chain reaction(FQ-PCR) methods in NP69-SV40T immortalized human nasopharyngeal epithelial, and 5-8F, 6-10B NPC cell lines.RESULTS MMP-9 was lower in NPC cell lines than in NP69-SV40T cell line (P<0.05), Of the two NPC cell lines 5-8F and 6-10B, the expression of MMP-9 mRNA was slightly higher in 5-8F, but no statistical significance was observed(P>0.05). RAGE mRNA was lower in NPC cell lines than in NP69-SV40T cell line and no statistical significance among the three groups(P>0.05). There were also no statistical significance among the three groups in expression of RECK mRNA(P>0.05).CONCLUSION There was no significant correlation between RECK, RAGE, MMP-9 and invasion, metastasis of nasopharyngeal carcinoma in vitro. OBJECTIVE To investigate the expression and the relativity of RECK,RAGE and MMP-9 protein in nasopharyngeal carcinoma (NPC) tissues.METHOD RECK,RAGE and MMP-9 were detected with immunohistochemiscal methods using antibody in sixty-four NPC specimens(30 cases with neck lymph nodes metastasis, while the other 34 cases are not) and 30 specimens with chronic nasopharyngitis.RESULTS RECK hardly expressed in chronic nasopharyngitis and NPC tissues, and there was no significant difference among the three groups (P>0.05), However, strong expression of RECK could be seen in the surrounding inflammatory cells and matrix of cancer nests. The postive rate of RECK in NPC with neck lymph nodes metastasis and otherwise was 3.3%(1/30) and 11.8% (4/34) separately, There was no statistical significance between the groups (P>0.05). The postive rate of RAGE in chronic nasopharyngitis and NPC tissues was 100%(30/30) and 70.3%(45/64)respectively, while MMP-9 was 46.7%(14/30)and 78.1% (50/64) . The postive rate of RAGE in NPC with neck lymph nodes metastasis and otherwise was 86.7%(26/30) and 55.9% (19/34) separately, whereas MMP-9 was 90.0%(27/30) and 67.7%(23/34) respectively. They all showed statistical significance between the groups (P<0.05). MMP-9 had a negtive correlation with RECK (r=0.471, p<0.05) and a postive correlation with RAGE in NPC(r=-0.369, p<0.05).CONCLUSION The expression of RECK and RAGE in NPC was down-regulated, while the expression of MMP-9 was up-regulated. The expression of RECK in NPC with neck lymph nodes metastasis was down-regulated, whereas RAGE and MMP-9 was up-regulated. The abnormal expression of the three genes may be related to the progress of NPC. RECK, RAGE may be involved in the process of invasion and metastasis of NPC by regulating the expression of MMP-9.
