Objectives:To detect the expression of HSF1in the human tissues of HCCand human HCC cell lines. And to investigate the expression of HSF1andrelationship between HSF1expression and human HCC cell proliferation andmetastasis. To achieve early diagnosis of HCC at the genetic level and toimprove the prognosis of HCC patients.Methods: The expression of HSF1gene in40samples of HCC tissue,40samples of adjacent tissue and10samples of normal liver tissue was detectedusing real-time PCR and immunohistochemistry. The expression of HSF1genein MHCC97-H cell line and MHCC97-L cell line was detected using real-timePCR and Western blot. The growth and cloning proliferation ability ofMCC97-H cells, MHCC97-H/HSF1-siRNA cells and MHCC97-L cells wasdetected using MTT and Flat cloning formation experiment.Results: 1. The HSF1mRNA expression level in HCC tissues was significantlyhigher than that in the adjacent tissues and normal liver tissues. The positive rateof HSF1protein in hepatocellular carcinoma tissues was significantly higherthan that in adjacent tissues, there was immunohistochemically negative innormal liver tissue.2.There was a positive correlation between HSF1protein and HSP70protein expression in HCC tissues.3.The high expression of HSF1protein was correlated with the integrity ofHCC’s envelop, cell differentiation and TNM staging, but it was not associatedwith the age, gender, HBV infection, AFP level, and phlebocarcinoma in portalvein.4.The HSF1gene expression level in MHCC97-H cell line wassignificantly higher than that in MHCC97-L cell line.5. The growth and cloning proliferation ability of MHCC97-H/HSF1-siRNA cells was significantly lower than that of MHCC97-H cells. Thegrowth and cloning proliferation ability of MHCC97-H/HSF1-siRNA cells wassignificantly higher than that of MHCC97-L cells.Conclusion: The above experiments suggested that HSF1may be closelyassociated with the development, advancement, invasion, metastasis andmalignant prognosis of HCC, perhaps it accomplishes these effects byregulating the expression of HSP70. HSF1, to some extent, also involved in theregulation of the growth and cloning proliferation ability of HCC cell lines...
