Most pants in China grow wild in the alpine zone and adjacent regions of Sichuan,Guangxi,Yunnan,Chongqing at elevations of 3200-3800m. Different extracts of this plant have been found to exhibit interesting properities such as anti-inflammation, anti-cancer,immunomodulation, free radical scavenging and anti-fatigue. Dried root of S. Lappa has been traditionally used for alleviating pain in abdominal distention and tenesmus, indigestion with anorexia, dysentery, nausea, and vomiting.But the research on treatment of Diabetea and Its syndrome isn’t appeared in the pharmacological study. In order to develop the natural resources,enrich the diversities of Traditional Chinese Medicine and find out new compounds from the root of Saussurea Lappa,we use the Pharmacologic model to screen of S. Lappa constituents with hypoglycemic activity.The research results as follow:1、Extraction and purification of the active position from S. Lappawe use the technologies of Silica gel column chromatography, Solvent extraction to analyse the active substance of S. Lappa. The results show, there are many hypoglycemic ingredients in the ethyl acetate extraction. We use the technologies of Silica gel column chromatography to deal with the extraction.We find the major compounds of costunolide and dehydrocostus lactone which is the hypoglycemic substance in the extraction.2、The study of antioxidant activity about S. LappaResearch on the Antioxidation of rhizome’s active siteBased on the diabetic mice model, hypoglycemic actiona-glucosidase, HepG2cells, mitochondrial model showing the results:costunolide and dehydrocostus lactone could significantly increase SOD activation and decreasing MDA content,decreaseing the ROS content of the cell and the mitochondria.It is the major active consisition of the antioxidation3、The study of antioxidant mechanism about S. LappaWith mitochondria which extract from rat liver as experimental,disturbing effect on membrane stability and fluorescence quenching effect of the costunolide and dehydrocostus lactone on mitochondrial nenbrane protein were determined by fluorescence spectrum analytical method.The results shows that they all can effect the stability of mitochondrial nenbrane protein. They all can quench the membrane protein.
