Studies On Technology For Extracting Costunolide

OBJECTIVE： To optimize extract technology for costunolide from root ofAucklandia lappa.METHODS：Based on the results of preliminary screening test and several singlefactor screening tests.The ethanol extract technology was chosen and optimized by anorthogonal test.The ethanol concentration，ratio of ethanol to drug materia，extractiontime and numbers were examined at three levels and a multi-target grading methodwas applied，which synthesized the contents of costunolide and the yield of dryextract.RESULTS：Ethanol concentration significantly effected on multi-target grade ofthe contents of costunolide and the yield of dry extract.The significant of ratio ofethanol to drug materia effected on multi-target grade of the contents of costunolideand the yield of dry extract less than that of ethanol concentration.The extraction timeand numbers could be selected in accordance with production practice.On the basis ofa multi-target grading analysis，The best method for extracting costunolide from rootof Aucklandia lappa is obtained as the herb is extracted with4folds aqueous ofethanol solution（85%）for three numbers and each extract last2h by an orthogonaltest.CONCLUSION：Compared with the original extraction process to optimize theextraction process，dry extract yield（extraction ratio）of Radix Aucklandiae andCostunolide transfer rate are increased，and provide evidence and guidance for thestudy of optimal production of Changweishu Capsule.
