Objective To explore the effects of antibiotic on intestinal flora and anastomotic bursting pressure in partial colectomy Wistar rat.Methods60Wistar rats were randomly divided into three groups:All the rats were underwent partial colectomy. During the perioperative period, group C rats (control group) were not received antibiotic at all, group A rats (Single antibiotics group) were received antibiotic once before the operation0.5hour prophylacticly, group B rats (Multiple antibiotics group)were received antibiotic until4days after the operation.7days after the surgery, all survival animals were killed. Anastomotic Bursting pressure were measured within5minutes. During the surgery and when the rats were killed, the colonic content was collected from the anastomotic stoma for flora analysis. And the16srDNA high throughput sequencing method was used for the flora analysis.Results After Illumina PE sequencing of the12samples, we got about4.12Gb raw data. Through analysis, we got that compared wth the preoperational group, the Shannon and npShannon index was lower than that in the three postoperational groups, while Simpson index was higher. Shannon and npShannon index was lowest and Simpson index was highest in the group B. The difference in the four groups was contributed mainly with Bacteroidales, Aeromonadales, Lactobacillales, and Clostridiles to the control group. The anastomotic bursting pressure was significantly lower in Multiple antibiotics group (p<0.05), and there was no significantly differences between the control group and the Single antibiotics group(p=0.961>0.05).Conclusions the antibiotic abuse can lead to the flora alternation, even dysbacteriosis, and these can result in a delayed healing, even anastomotic dehiscence.
