Objective: To explore the structural characteristics and differential analysis of intestinal flora in obese children and normal weight children,and lay a foundation for further study on the prevention and treatment of obese children.Methods:Feces were collected from 30 obese children in the outpatient department of the second affiliated hospital of anhui medical university and 30 normal weight children in a primary school in anhui province.16 Sr DNA detection method was used for DNA extraction and detection,PCR amplification,product purification,library preparation and library detection.Ion S5 TMXL was sequenced on the machine,and a small fragment library was constructed for terminal sequencing by single-end sequencing.By cutting and filtering,reading,OUTs clustering,and finally species annotation,abundance analysis and diversity analysis,the composition of intestinal microorganisms and the differences between intestinal flora are understood.Results:In the OUT annotation,the average number of clean reads in obese and normal-weight children was 79963.3 and 80860.7,indicating that fewer sequences were ultimately used for subsequent analysis in obese patients than in the normal-weight group.In Alpha Diversity,there was no significant difference between the indexes of Chaol and ACE,indicating that the species richness of children in the obese group and the normal weight group was consisten.The species composition of the top ten species in phylum,class,order,family and genus in the obese group and the normal weight group was basically similar,but the proportions were different.At the phylum level,Firmicutes,Bacteroides and Actinomycetes are the most abundant flora in the intestinal microbial community of children.Among them,Verrucomicrobia and Acidobacteria were significantly higher in normal weight group than in obese group.The level of Erysipelas in class,order and family was higher in the obese group than in the normal weight group.At the level of genus,Akkermansiaceae?Massilia?Gammaproteobacteria?Candidatus-Kolibacter,Akkermansiaceae,Bradyrhzzobium are found that the children in the normal weight group were significantly higher than those in the obesity group.Dorea was significantly higher in the obese group than in the normal-weight group.At the specific level,Bacterodies-thetaiotaomicron,Clostridium-scindens were found that the children in the normal weight group were significantly higher than those in the obesity group.Dorea-longicatena,Acteroides-massiliensis and Clostridium aeruginosa were significantly higher in the obese group than in the normal weight group.Conclusion: The species richness and diversity of intestinal flora of obese children and normal weight children were analyzed by 16 Sr DNA,and it was found that the two groups were consistent.The distribution and quantity of intestinal flora affect the occurrence of obesity.
