Purpose:1)Using High throughput sequencing technology to analysis the structural of intestinal flora in the breast cancer patients of Uygur and Han nationality and corresponding healthy women.To study the structure and diversity of the intestinal flora in Uygur and Han patients with breast cancer.To explore the possible correlation between intestinal microflora and breast cancer.To construct a preliminary study on the composition of intestinal microflora in different nationalities of breast cancer,and to screen out the strains which are significantly related to the disease.2)Analysis of the difference of the intestinal flora in the Han nationality women with breast cancer and breast cancer patients in Uyghur people by using the same method.Preliminary study on the diversity and structural similarities and differences of intestinal flora in two women with breast cancer.Method:Illumina Miseq platform was used to carry out high throughput sequencing of 16SrDNA V3-V6 region in the Han and Uygur women with breast cancer and their corresponding healthy women,Sequencing resuLts were analyzed by OTU cluster analysis,diversity and structure analysis.The distribution,abundance and diversity of intestinal flora in two nationalities were obtained.Result:1)There was no significant difference in the mean OTUs number between OTUs Uygur breast cancer?n=10?and healthy controls?n= 10??P>0.05?;The mean number of OTUs?n=25?in the Han Chinese breast cancer group was significantly lower than that in the healthy control group?n=15?;The complexity analysis by dilution curve,Chao1 index,Shannon index and Rank abundance curves for four samples,sequencing amount sufficient to cover intestinal bacterial species,richness and evenness of each sample was better.2)At the genus level,there were significant differences among the Uygur women with breast cancer and the healthy group of Bacteroides,Lachnospira and Alicyclobacillus?P<0.05?;There were significant differences between the 13 groups of the genus SubdoligranuLun Alistipes,Blautia,and so on in the Han nationality women with breast cancer and health group?P<0.05?;There was a significant difference in breast cancer group and Han breast cancer group in the genus is prevolla9,proposed 19 kinds of bacteria Bacillus including Succinivibrio,SubdoligranuLum,Parabacteroides,Ruminococcus2 and other genera?P<0.05?;There were significant differences between the normal group of Uyghur women and the normal group of Han nationality,including 7 genus?P<0.05?,which belong to the genus,such as Bacteroides,Megamonas,Megasphaerea and Bifidobacterium.3)There were differences in the structure of the intestinal flora between the group of Uygur women with breast cancer and healthy women?Chinese women with breast cancer and healthy women?women with breast cancer in Han and Uygur women with breast cancer?Han nationality and the Uygur women,And each group has its dominant species at each level.Conclusion:1)There were significant changes in the structure of intestinal flora between the two breast cancer patients and healthy controls,It is suggested that the changes in the structure of the intestinal flora may be associated with the disease status of breast cancer.2)There are differences in the composition of the intestinal flora between the Han Chinese women with breast cancer and the Uygur women with breast cancer,This difference may be the resuLt of a combination of genetic and environmental factors.3)There were significant differences in the proportion of intestinal flora in healthy Han and Uygur women,The resuLts may be related to genotype and diet.
