Objective1.By understanding the impact of IL-10、TNF-αon rat placental lactogrn, to discuss the influence of Th1 / Th2 Type of cell factors in the role of placental lactogrn secretion.2.To investigate when pregnant mice serum TNF alpha level rises, how the pregnant miceserum levels of mice PL will change,if giving IL-10 to intervene.MethodsPut sexual maturity mice into one cage at each day’s 6:00 pm to pass the night,accordingto the proportion of male and female 1:2, and at 8:00 the next morning observed the Yin bolt.Recorded mice that had Yin bolts as pregnancy 0 day , and marked pregnancy mice.Then allsixty pregnancy mice were divided to six groups randomly. The first group was salinesolution group,the second group was 20ng TNF-αgroup,the third group was 50ng IL-10group,the fourth group was 20ng TNF-α+50ng IL-10 group,the fifth group was 20ngTNF-α+100ng IL-10 group,and the sixth group was 20ng TNF-α+200ng IL-10 group.Onday7,8, 9,10,11 of gestation,the first group mice were injected with saline solution byabdominal, and other 5 groups mice were injected different levels of IL-10、TNF-αintraperitoneally.On day 12 of gestation,the mice were killed and detected the serum levels ofmice PL by enzyme linked immunosorbent assay(ELISA).Compared the differences of theserum levels of mice PL with analysis of variance and Kruskal-wallis.Results1.The serum levels of mice PL in the second group was significantly higher than the first group and third group(P<0.01), but the serum levels of mice PL in the third group had noapparent changes compared with the first group( P>0.05).2.The serum levels of mice PL in the second group、the fourth group、the fifth group and thesixth group had no significant changes ( P>0.05).In four groups serum mice PL mean visible, the latter three groups of serum mice PL level were lower than 20 ng TNF alpha group.Conclusion1. In this lab, TNF alpha group significantly increased the serum level of mice PL comparingwith the physiological saline group (a normal pregnancy group), concluded that TNF alphahad the function of promoting placental lactogrn secretion.2. IL-10 had no significant function on pregnant mice PL secretion. 3. When pregnant mice serum TNF alpha level significantly increase, given this lab doses ofIL-10 intervention, the serum levels of mice PL had no significant change.
