Objective:To explore the alterations of activity,antigen and genotypes of the thrombin-activatable fibrinolysis inhibitor in VTE.Methods:80patients that including12PTE patients and68DVT patients,80controls were studied. The thrombin-activatable fibrinolysis inhibitor (TAFI:Ac and TAFI:Ag) was measured among the patients and controls using ELISA assay. The TAFI505A/G and1040C/T SNP genotype in the two groups were determine by AS-PCR.And by sequencing we certified this results.Results:1.The plasma level of antibody and activity was elevated significantly in VTE patients than in controls.2. The plasma level of Antigen has obvious relationship with the AA genotype of505A/G,but the piasma level of Antigen has no relationship with the1040C/T.3.The distribution of genotype was calculate by HWE equilibrium. The frequencies of TAFI505A/G have disparity,but they were not statistics differences. The frequencies of alleleC were60%vs25%, alleleT were40%vs53.75%.The allele frequencies and genotype has no statistic differences.The frequencies of CC genotype of1040C/T were33.8%vs18.8%in the VTE group and control group, CT were52.5%vs55%, TT were13.7%vs26.2%respectively.4.The plasma level of TAFI:Ac has no relationship with the TAFI505A/G and1040C/T genotypes between the two groups(p>0.05).Conclusion:1.It suggest that the activity and antibody may be play the important rule in VTE group.2.The polymorphism has no relationshiop in505A/G between the two groups. But there was obvious significant relationship in the polymorphism of1040C/T in TAFI between the normal controls and VTE group,T allele genotype may be paly a protect role in VTE.3.The antibody plasma level of TAFI has significantly relationship in the505AA of the TAFI,but the piasma level of Ag has no relationship with the TAFI1040C/T.4.The plasma level of TAFI:Ac has no relationship with the TAFI505A/G and1040C/T genotypes in the two groups.
