Ependymoma tumor is rare,accounting for3%of all intracranialneoplasms. Intracranial ependymomas arised from the ependymal cells ofbrain wall and the surface of spinal cord central tube. Intracranialependymomas generally occur in children.Spinal cord ependymomas,howeverare seen more frequently in adult patients. The Characteristic pathologicalperformance of ependymoma is c ependymal rosettes and perivascularpseudorosettes. Ependynlomas continue to generate considerable conortversyregarding their rational clinical management. With the development ofmicro-surgery technique it is widely recongnized that surgery is the mainstay ofmanagment. That glioma invasion and recurrence is the result of a variety offactors regulation such as genes, proteins and cytokines. This process mayinvolve tumor cell adhesion, matrix degradation, migration, cell proliferationand many other areas. How to explore the glioma cells of early invasion andmetastasis mechanism from the perspective of molecular biology, provide atheoretical basis for early intervention to block glioma invasion, is one of thefocus of cancer gene therapy.Haptoglobin is one of acute-phase acidic glycoprotein in the serumglobulin fraction and widespread in humans and many mammals, serum andother body fluids.Hp is mainly synthesis and degradation in the liver. As one ofthe acute phase protein, its serum levels were often significantly higher Whenthe body is in stress or serious illnesss such as cancer, inflammation, trauma,infection, myocardial infarction and other pathological conditions.and it isrelative to the severity and prognosis. Fibrinogen (Fg) is clotting factor I andthe precursor of fibrin, is an asymmetry high molecular weight protein. The main place of synthesis and secretion is Liver.The final stages of coagulation isthe formation of thrombin, in this stage soluble fibrinogen transforming intoinsoluble fibrin and blood coagulation. Fg Is also an acute phase protein,andIts increasing is common in cholecystitis, light hepatitis, pneumonia,tuberculosis and other infections and malignant tumors, cerebral thrombosis,cerebral infarction, rheumatic fever, aseptic inflammation Fibrinogen directlyinvolved in the clotting process,and blood viscosity,is an important risk factorfor heart and cerebrovascular disease. The levels of plasma fibrinogenincreased in patients with malignant tumor, comparing with the normal diseaseand benign tumors,it often significantly increased,The relevant reports aboutHp and Fg is rare at home and abroad, Dr. Li Wenchen have confirmed in thepaper that named 《Comparative proteomic study of cerebrospinal fluid inbetween patients with medulloblastoma and patients with ependymoma》thatcomparing with normal control group and the medulloblastoma group,Hp andFG were significantly increased in the ependymoma group,howevermedulloblastoma group and normal control group had no significantdifferences.The aim of this study is to investigate the the expression of Hp,Fg byimmunohistochemistry two step method and their correlations withclinicopathologic features of ependymoma in order to explain the mechanismof malignant behavior.Materials and methods:1.clinical data:10ctissue samples operated in the NO.1hospital of Jilin University fromMarch.2010to August.2011were collected.Together with10cases ofnon-tumor brain tissues,taken from operated patients with brain trauma orhypertensive cerebral hemorrhage were studied as control group.All tumorswere verified and cateogorize by pathologist in line with WHO(1999) criteria for ependymoma.All samples were10%formalinfixed,and para-ffinembedded,and cut into3-4um slices to be examined.2.Immunostaining:Hp and Fg monoclonal antibody were purchased from Beijing Bao’ao’senbiological techonology company. SP-9710and DAB kit was purchased fromFuzhou Maixin biological techonology company.3.Determination:Image analysis use Motic Images Advanced3.2Image analysis software.Hp and Fg are all scheduled in the cytoplasm. Cytoplasm appered brown whichwas identified as postive cell. In the field of vision of200times, selected fivefields of vision randomly, measured gray scale value. Depending on the size ofthe gray value, calculate the staining intensity,the smaller of the measured grayvalue, the deeper of cytoplasmic staining,and the more of the content of themetachromatic granule of intracellular. Metachromatic granules is inverselyproportional to the gray value.4. Statistical analysis:All the data were treated with software “SPSS13.0”.t-test was applied instudying correlation between Hp or Fg and tumor clinicopathologiccharateristics,and the relationship between Hp and Fg was tested by means ofPearson correlation analysis.Results:The results of experiment demostrated that the expression of Hp and FGwas mainly localized in the cytoplasm.By t-test,P<0.05, the expression of Hpand FG in ependymoma and normal brain tissue is different,and has statisticalsignificance.we can perorate that the expression of Hp and FG inependymoma is higher than normal brain tissue,and prompt that the increasedexpression of Hp and FG is related to the development of ependymoma. ByPearson correlation analysis,comparing the expression of Hp and Fg in normal brain tissue with ependymoma,we found P>0.05,there was not statisticalsignificance between Hp and Fg.Conclusion:1.The expression of Hp and Fg in ependymoma is higher than normalbrain tissue,It prompt that Hp and Fg may play an important role in thedevelopment and invasive of ependymoma.2.The expression of Hp and Fg in ependymoma was not relevant,and stillcan not determine Whether Hp and Fg has a synergistic effect in the occurrenceand development of ependymoma.
