BackgroundTumstatin comes form noncollagen protein NC1structure domain in collagen Ⅳα3chian and contains244amino acid residues; At present, it is the most active endogenous tumor angiogenesis inhibiting factor as we known, at the same time it also can promate the tumor cell apoptosis straightforwardly and have a direct relationship with tumor formation and metastasis.Matrix metalloproteinases are a highly conservative in peptide enzymes which rely on calcium and zinc, it has been confirmed that they can degradation the extracellular matrix and further tumor neovasculature and play a important role in the tumor invasion and metastasis of embellish process.Up to now, it turns out that tumstatin can restrain the growths of cell strain of prostatic carcinoma and renal cell carcinoma and so on; It has been found that MMP-9has a high expression in tumor tissue.But, the expression of tumstatin and MMP-9in the prostatic carcinoma tissue and the relationship among clinical and pathologic stage and PSA correlated variates have not been reported.ObjectiveResearch the expression of tumstatin and MMP-9in prostatic carcinoma and benign prostatic hyperplasia.Explor the relationship between tumstatin、MMP-9and clinical and pathologic stage, also the origin and development of the prostatic carcinoma.MethodImmunohistochemistry was used which to detect the expression of tumstatin and MMP-9in68patients with prostatic carcinoma and42patients with benign prostatic hyperplasia.Result1. The positive expression rate of tunatatin between prostatic carcinoma and benign prostatic hyperplasia tissue is different。The positive expression rate of tunatatin in prostatic carcinoma tissue is38.2%less than66.7%in benign prostatic hyperplasia tissue.There is significant difference between them (X2=8.398, P=0.004<0.05). The positive expression rate of tunatatin in prostatic carcinoma patients whos age less than60is30%also it is39.7%in prostatic carcinoma patients whos age more than equal to60. There is nosignificant difference between them (X2=0.337, P=0.562>0.05).The positive expression rate of tunatatin in well-differentiated prostatic carcinoma tissue is83.3%, but, in moderately and poor-differentiated prostatic carcinoma tissue, they are29.2%and25%.There is significant difference among them (X2=12.414, P=0.002<0.05). The positive expression rate of tunatatin in clinical stage A、B、C、D prostatic carcinoma tissue are81.8%、36%、27.3%、23.8%. There is significant difference among them (X2=11.114, P=0.011<0.05)2. The positive expression rate of MMP-9between prostatic carcinoma and benign prostatic hyperplasia tissue is different。The positive expression rate of tunatatin in prostatic carcinoma tissue is80.9%less than28.6%in benign prostatic hyperplasia tissue.There is significant difference between them (X2=29.84, P=0.000<0.05). The positive expression rate of MMP-9in prostatic carcinoma patients whos age less than60is80%also it is81%in prostatic carcinoma patients whos age more than equal to60. There is nosignificant difference between them (X2 =0.009, P=0.939>0.05) The positive expression rate of MMP-9in well-differentiated prostatic carcinoma tissue is66.7%, but, in moderately and poor-differentiated prostatic carcinoma tissue, they are87.3%and87.5%.There is significant difference among them (X2=6.513, P=0.039<0.05) The positive expression rate of tunatatin in clinical stage A、B、C、D prostatic carcinoma tissue are45.5%、80%、90.9%、95.2%. There is significant difference among them (X2=18.699, P=0.000<0.05).3. In prostatic carcinoma tissue, the expression of tumstatin and MMP-9was negatively correlated (r=-0.310, P=0.011<0.05), FPSA and PSAD are positive correlated with pathological stage of prostate carcinoma but not correlated with clinical stage.F/TPSA is not correlated with pathological and clinical stage.ConclusionTumstatin and MMP-9were expressed in prostatic carcinoma and benign prostatic hyperplasia tissue。Tumstatin was lowly expressed in prostatic carcinoma but highly expressed in benign prostatic hyperplasia tissue, The positive expression rate of tumstatin in prostatic carcinoma related with the clinical and pathological stage but not related with the age of the patients. MMP-9was highly expressed in prostatic carcinoma tissue, The positive expression rate of MMP-9in prostatic carcinoma tissue related with the clinical and pathological stage but also not related with the age of the patients. Tumstatin and MMP-9may be markers for the diagnosis and treatment of prostatic carcinoma. The expression of tumstatin and MMP-9in prostatic tissue was negatively correlated; Tumstatin is expected to become the new research directin for prostatic carcinoma. In the serum prostate cancer patients, FPSA and PSAD are positive correlated with pathological stage of prostate carcinoma but not correlated with clinical stage.F/TPSA is not correlated with pathological and clinical stage, The PSA correlated variates ideal index for prostate cancer clinical work.
