Objective: The author carded out some experiments to assess the embolization effectand biocompatibility of CS/β-GP as intravascular liquid embolic materials.Methods:1.⑴We get the CS/β-GP by mixing Chitosn and Sodium Glycerophosphateat the ratio of7:1, then the CS/β-GP was injected into the1.8F,2.5F,3F,4F catheter insequence in water bath (PH7.4,37℃), and then we observed the viscosity.⑵The effects ofcontrollability and embolization for the CS/β-GP were evaluated by injection test andaneurysm embolization experiment in vitro.2.⑴We put the leach liquor of CS/β-GP gelas test solution. Mice were intraperitoneal injection of the leach liquor so as to observe thesystemic toxicity in mice.Each mouse of every group underwent intraperitoneal injectionwith leach liquor(50ml/kg), while the control group injection of0.9%sodium chlorideinjection(50ml/kg) for parallel control in the same period. The general condition of animalswere observed including appetite,weight change,activity, toxic reactions animal number.According to the toxic evaluation classification to evaluate toxicity reaction. and even thenumber of dead mice.According to the group,the mice were sacrificed in correspondingobservation period respectivel.The organs including brain,heart,lung,liver,spleen,kidneyand epiploon were taken respectively for general and histology analysis.⑵Add10ml leachliquor into test tube, bathe in37±0.5℃water for30minutes.0.2ml of fresh blood was addedinto it,shake even,bathe in37±0.5℃water for60minutes and centrifug,and then opticaldensity was measured on spectrophotometer at540nm.Finally, According to the formula tocalculate the hemolysis rate.⑶Leach liquor was intraperitoneal injected into each mouse,and0.9%sodium chloride injection(50ml/kg)for each animal in control group.Bleeding timeand coagulation time were detected after7d.Results:1. Material can be smoothly injected into these three types of microcatheter,including the2.5F,3F,4F catheter. Just not easy through the catheter of1.8F. Theaneurysm model was successfully embolized by CS/β-GP gel, and there were no materialadhere to cather after embolization.2.⑴During observation period,the general condition ofall experimental animals were well. Compared to the control group,In all specimens there was no cell degeneration and necrosis,no exudation and inflammatory cells infiltration.⑵The test hemolysis rate was3.9%, less than5%, according to GB/T16886standard, CS/β-GPcause no hemolysis as intravascular liquid embolic material.⑶The difference of thebleeding time and coagulation time between experimental group and control group wasstatistically significant(P>0.05), comply with the requirements.Conclusion: As intravascular liquid embolic material, CS/β-GP have good feasibilityand excellent biocompatibility.
