Objective:To investigate changes of kidney function in infants who consumed melamine-contaminated milk powder, and discuss the influence of melamine on kidney function; A novel flow-injection chemiluminescence method for the determination of melamine in urine and plasma was developed.Methods:1. From April to December in2008,180infants with positive urine were investigated in the action of "The survey of melamine-induced urinary stones in infants in Gansu province". Urinary albumin, β2-microglobulin and a1-microglobulin were determined quantitatively so as to observe whether melamine results in long-term injury to the kidney, as well as the correlation between the injury and stone site, gender, age, duration of milk consumption, respectively.2. Under alkaline conditions, melamine can significantly strengthen luminous intensity of luminol-K.3FeCN6system, meanwhile the system is relatively stable. So the flow injection chemiluminescence method for determination of melamine was established.Results:1. The results showed that there were31cases with abnormal urinary albumin and19of180cases with βVmicroglobulin abnormalities, but the index of a1-microglobulin are normal. Urinary albumin is statistically different between two groups. The abnormal level of UMA and β2-MG has positive correlation with milk usage.2. Under the optimum conditions, chemiluminescence intensity had a good linear relationship with the concentration of melamine in the range from9.0×109to7.0×106g/mL with a correlation coefficient of0.9992. The detection limits was3.5ng/mL. The method has been applied to determine the concentration of melamine in the samples with a liquid-liquid extraction. Average recoveries of melamine were102.6%in urine sample and95.1%in plasma sample. Conclusion:1. Melamine may caused injury to kidney glomerular and tubular function. The more consumption of milk powder the severe injury to kidney function.2. A novel flow injection chemiluminescence method was established to determine melamine content in blood and urine.
