Liucha, which used the leaves and twigs of Sibiraea laevigata(L.) Maxim. or S. angustata (Rehd.) Hand.-Mazz., is a plant of Sibiraea in Rosaceae. Tibetans use it to calenture and loemia, to resolve food stagnation and Dispel wind-heat. Liucha is mainly distributed in Gansu, Qinghai, Sichuan and Xizang. In recent years, with the increased pharmacology study of Liucha, the effect on adjusting lipid metabolism, antioxidation, promoting digest, improving immunity and reducing blood sugar of fatty rats of Liucha have been found. It has good advantage for development because of its rich resources, sustainable development. On the basis of collection the data of Chinese herbal medicines in Gansu province, we make a research of Liucha on diabetic mice induced by Alloxan. Then we established a determinational method with GC-MS to free fatty acids in serum. We study the influence of extraction of Liucha and compound(Liucha, Astragalus, Rosa) on free fatty acids in rats with hyperlipemia.Our group observed the effect of water-extraction of Liucha on Diabetic mice induced by Alloxan, including blood glucose, sugar tolerance and the pathological changes of pancreas. We established the diabetic rat model with Alloxan(ALX). We divided them into five groups, Which were fed with distilled water. Xiaokeling, different dosage of extraction of Liucha (high:2.90g·kg-1·d-1; middle:2.03g·kg-1·d-1; low:1.16g·kg-1·d-1). The two nomal groups were separately fed with distilled water and high dosage of Liucha. Middle and low dosage groups can make the blood sugar of Diabetic mice decreased and were benefit for sugar tolerance; Hige dosage of Liucha can moderate the curve of sugar tolerance, but have no effect on sugar; they have no effect on sugar of nomal mice; High and middle groups can make the lesions of pancreas recurred to a certain degree. The mechanism is maybe related to Liucha’s repairation for β-cells of pancreas and its metabolic adjustment.We established a method with GC-MS to analyse free fatty acids, such as Nonanoic acid, Dodecanoic acid, Tetradecanoic acid, Hexadecanoic acid, Heptadecanoic acid, Linoleic acid, Oleic acid, Stearic acid, Arachidonic acid, Eicosapntemacnioc acid and Arachidic acid, at the same time. This method has good linear relationship, precision and repeatability, RSD is always within10%, sample recovery is between80%and120%. We use this method to study the effect of water-extraction of Liucha and compound on hyperlipidemia rats. We divided hyperlipidemia rats into8group and black group. We collected blood with method of abdominal aorta, then determinated the content of free fatty acids in serum. As a result, the effect of water-extraction of compound is stronger than Liucha. The crude polysaccharide has no effect. The method is also adapt to other study on Cardiovascular and metabolic disease.
