Part1hENT1mRNA expression in relation to prognostic of Non-small cell lung cancer patients received gemcitabine-based chemotherapy Aim of this study was to investigate the correlation of the mRNA expression levels of human equilibritive nucleoside transporter1(hENT1) with clinicopathological parameters in patients with non-small cell lung cancer receiving gemcitabine-based chemotherapy as well as their clincal prognostic significance. The relative mRNA expression of hENT1to the internal reference gene β-actin in formalin-fixed paraffin-embeded non-small cell lung cancer tumor specimens was measured using real-time quantitative reverse transcription polymerase chain reaction.And the relationship between hENT1levels with clinicopathological parameters and survival in non-small cell lung cancer was studied. hENT1expression levels were detected in all tumor specimens.hENTl expression was not related to clinicopathological parameters in non-small cell lung cancer. Though from multivariate analysis, hENT1mRNA hypoexpression was not a significantly favorable factor for overall survival(P=0.707),and for time to progression(P=0.420), there is a tendency that patients with high hENT1mRNA levels had a significant longer median time to overall survival and progression. Although from Cox proportional hazards model for survival, hENT1mRNA hypoexpression was not a significantly favorable factor for OS and PFS,tumor lymph node matastasis was an independent predictive marker of overall survival of non-small cell lung cancer.(P=0.02).It was suggested that intratumoral hENT1mRNA expression level,not associated with clinicopathological parmeters,maybe a predictive marker of survival of gemcitabine-containing combination chemotherapy in non-small cell lung cancer.It can provide critical information for planning personalized chemotherapy with a larger sample size. Tumor lymph node matastasis was an independent predictive marker of survival of non-small cell lung cancer.
