Objective:To investigate significance of ribonucleotide reductase M1(RRM1) expression in stage ⅢB~Ⅳ non-small cell lung cancer(NSCLC) predicting gemcitabine-based chemotherapy efficacy and provide proper medication for patiens.Methods:From February2010to February2012,patiens in stages IIIB~IV non-small cell lung cancer patiens were diagnosed by pathology in the way of video bronchoscope and percutaneous lung puncture then40patiens were collected in oncology department of the affiliated hospital of Chengdu university. The RRM1expression in tumor samples was detected by the immunohistochemical technique.The patients were divided into positive RRM1expression group(20patients) and negative expression group(20patients) according to their expression level of RRM1.They both were given gemcitabine-based chemotherapy2cycles.The chi-square test is used to compare the sample rate; The chi-square test and the rank sum test were used to compare the RRM1expression correlation with the curative effect in the near future.Kaplan-meier is used to compare the RRM1expression correlation with TTP and analyse one-way survival. Log-rank is used to test the result.Results:There was no correlation between the RRM1positve expression situated in the advanced non-small cell lung cancer tissue and clinical characteristics such as gender,age,smoking,pathological type and TNM stage. Chemotherapy efficacy (CR+PR) of patients in the RRM1positve expression group is15.00%(3/20); otherwise RRM1negative expression group is50.00%(10/20)(P<0.05).Chemotherapy efficacy of the RRM1negative expression group is better than those of the RRM1positve expression group,which is statistically significance (P<0.05). The medial TTP was88days in the RRM1positve expression group,which was146days in the RRM1negative expression group;(P<0.05). The medial TTP of the RRM1negative expression group is better than those of the RRM1positve expression group;The TTP curve of the RRM1negative expression group is above those of the RRM1positve expression group, which is statistically significance.Conclusion:The RRM1expression level in tumor cells is a predictive marker of efficacy in advanced NSCLC patients received gemcitabine-based chemotherapy.
