Ilex pubescens Hook.Et Arn.is a plant of Aquifoliaceae Ilex L.which is traditional Chinese medicial, there are rich in natural resources which maily growing in GuangDong, GuangXi, AnHui, FuJian, ZheJiang, et al. Its root and leaves are used as medicial which can clearing heat and detoxicating, promoting blood circulation and removing obstruction in channels and so on. It is used for old, tonsil, dysentery, mouth disease and coronary disease. According to reports, the chemical constituents and pharmacology activity of Ilex pubescens root have been studied its chemical constituents and Systematicly, but the mainly studing on leaves is that use as medicial by formula, Its chemical constituents and pharmacology activity have been reported lees. Ursolic acid and Oleanolic acid has been extracted from Ilex pubescens leaves in1956, Ursolic acid,3,4-Dihydroxybenzoic acid and3,4-Dihydrox-ybenzaldehyde’s content of Ilex pubescens leaves has been analysed in1992.and the report that the content of ilexgeninA’s from Ilex pubescens leaves has been studies in2011. In this paper, we study the chemical constituents of Ilex pubescens leaves. In order to know the pharmacology of Ilex pubescens leaves in these formula, and found the inportant constituents with nice pharmacological activities. Fully utilizing this resources to respond the scientific idea of reduce resources.It include six parts in this paper. The first part was the review of the pesearch progress of Aquifoliaceae Ilex L. plant. The second part was studying the antioxidant activity of Ilex pubescens leaves’s extract with alcohol by UV spectrophotometry.The third part was introduced the Volatile oil from Ilex pubescens leaves which identified by GC-MS. The fourth part was studying the constituents by extraction, separate, purification and identified. The fifth part was studying the extraction and purification process of total triterpene acids and flavonoids of Ilex pubescens leaves. The last part was summarizing up the study of all experiments.The main results are show as following:(1) The antioxidant activity of30%,50%,70%,90%alcohol extract and two compounds of Ilex pubescens leaves were determined by UV spectrophotometry. The50%alcohol extract and90%alcohol extract showed obvious antioxidant effect. The result showed that at the same concentration,the efficiency of DPPH scavenging as following:Vc>50%>30%>90%>70%> A1> A2;the efficiency of O2scavenging as following:Vc>90%>A2> A1>30%>50%>70%; The efficiency of·OH scavenging as following:90%> Vc> A1> A2>50%>70%>30%. From this, the flavone of Ilex pubescens leaves has antioxidant capacity.(2) In this paper, the Volatile oil from petroleum ether and chloroform extract of Ilex pubescens leaves which identified by GC-MS, and compare with difference of them. The result show that the Volatile oil from petroleum ether extract mainly Contain hydro carbons (13.063%), phenolic compounds (33.624%), esters(15.587%), and the relative highest content compounds was (E)-2-methoxy-4-(1-propenyl)-Phenol (28.967%).he Volatile oil from chloroform extract mainly Contain hydro carbons(64.033%), Fatty acid(12.891%), the relative highest content compounds was Eicosane(8.735%).(3)22compounds were isolated and purfied from the alcohol extract of Ilex pubescens leaves, and16compounds were identified by spectrum technology, the16compounds were ChaenomelosideA (1),23-aldehydepolomic acid (2), Ursolic acid (3), IlexgeninA (4), Ilexolic acidB (5), Chaenomelog-eninA (6), β-daucousterol (7), Oleanolic acid (8), Hexacosane(9), β-sitosterol (10), Stigmasterol (11), Pentadec-anoic acid (12), Taraxerol (13), Hydroqui-nine (14), ChaenomelosideA (1),23-aldehydepolomic acid (2), ChaenomelogeninA (6), Taraxerol (13) were not reported in literature about Ilex pubescens.(4) The extraction process of triterpenoids from Ilex pubescens Hook leaves, The influence factors of extracting temperature, radio of liquid to material, extracting time and concentration of ethanol on the triterpenoidsof yield was evaluated by single factor test. The results showed that the optimum conditions were1:16in radio of liquid to material, concentration of ethanol70%and the extraction time90min at56℃. The extraction rate of triterpenoids was6.4868%.Then purified by D101macroporous resin, Its structure was characterized by IR spectrum, the result show that the total saponins has a same aglycone with Ilexgenin A.(5) The Content of total flavonoids of Ilex pubescens leaves was studying by NaNO3-Al(N O3)-NaOH and UV spectrophotometry, The influence factor of extracting temperature, radio of liquid to material, extracting time and concentration of ethanol on the flavonoids of yield was evaluated by single factor test. The results showed that the optimum conditions were temperature60℃, radio of liquid to material1:16, ethanol concentration80%and extraction time2h.
