The traditional Chinese medicine, Maodongqing, dry roots of Ilex pubescens Hook.et Arn. of Aquifoliaceae, has been used against such diseases caused by virus and fungi, even other usages in China. Coumarins, terpenes, amino acids, sterols, polysaccharides and some other types of constituents have been found in Maodongqing, and less than 35 compounds were determined clearly. In this paper, the chemical components of Maodongqing were furtherly stud- ied and its chemical compositions were added than before, it is very useful for development resource, exploration active components and accumulation of natural chemical information.The 95% EtOH extract of Maodongqing was re-extracted with petroleum ether, ethyl acetate, and water successively to give three fractions, among of which, fraction petroleum ether and ethyl acetate were isolated and purified, and thirteen compounds were separated and deter -mined from fraction petroleum ether and ethyl acetate, including daucosterol (6),tortoside A (7), liriodendrin (8), 1-hydroxypinoresinol-1-β-D-glucoside (9), Ilexgenin A (10), Oleanolic acid (11),β-sitosterol(12), Stigmastero1(13), Hydroquinone (14), Umbelliferone (15), chlorogenic acid (16), protocatechualdehyde (17), Succinic acid (18). Meantime, The 70% EtOH extract of Maodongqing was chromatographed on a D-101 macroporous resin and silica gel for several times, six triterpenoid saponins were obtained, and five of them were identified, including Ilexoside O (1), Ilexsaponin B3 (2), Ilexsaponin B1 (3), Ilexsaponin B2 (4), Ilexgenin A (5). Compounds 6, 9, and 12~18 were first isolated from Ilex pubescens Hook. et Arn.The review on the study of chemical constituents and pharmacological activities have been made. The molecular structure of some triterpenoid saponins , was elucidated by 1D NMR and 2D NMR techniques ( 1H , 13C , DEPT , COSY , TOCSY , HSQC , HMBC ) for the first time, and its complete assignment of 1H and 13C NMR chemical shifts were also obtained. In particular , the analytical method of combining 1H - 1H COSY and 1H- 1H TOCSY was applied in order to analyze overlapped H NMR signal of the sugar in detail , and a way to assign NMR signal of the sugar of triterpene saponin completely was established. All these work established foundation stone for the scientific research work of Ilex pubescens Hook. et Arn..
