Objective:Studying the effect of sinomenine hydrochloride on sperm quality and testis morphology of antisperm antibody immune sterility rat.Methods:The antigen was prepared by iso-SD rat sperm. Using artificial active immunization method to establish animal model of immune sterility of antisperm antibodies. The level of antisperm antibody (AsAb) in rat serum was determined by enzyme-linked immunosorbent assay (ELISA). The administration of drugs after the model was success. Observing sperm density, sperm survival rate, sperm viability. activity of sperm acrosin of each group rats after administration of the drug, and compared.RESULTS:1. In the model experiment, model group compared with the normal group, AsAb in rat serum concentrations increased significantly(P<0.05), and each model group between serum AsAb concentrations had no significant difference(P>0.05).2. Compared with the model group, after the treatment of the sinomenine hydrochloride, rat AsAb was remarkably reduced(P<0.05,or P<0.01),and the high dose group curative effect obviously(P<0.01).Low dose group although had treatment effect but poor,compared with the normal group, the serum AsAb concentration is higher(P<0.01).3. Compared with the model group, after the treatment of the sinomenine hydrochloride,rat sperm density was significantly increased(P<0.01).Sperm acrosin activity increased, but without statistical significance(P>0.05).Sperm activity rate, survival rate was significantly increased(P<0.05,or P<0.01).And the dose group had obvious effect(P<0.01).4. Compared with the model group, after the treatment of the sinomenine hydrochloride, grade B sperm number increased significantly(P<0.01), grade D sperm number decreased significantly(P<0.05,or P<0.01), and the high dose group effect obviously (P<0.01). Grade a+b sperm number increased significantly(P<0.01), Grade c+d sperm number decreased significantly(P<0.01).5. Compared with the model group, after the treatment of the sinomenine hydrochloride, seminiferous tubules、spennatogonium、 primary secondary spermatocytes of rat testicular are clear、orderly、 distinct. The each grade spermatogenic cell growth is good and the mature sperm is more. Testicular interstitial cell growth is good, no abnormal phenomenon.Conclusion:1. The normal group compared with model groups and’ model groups compared with each other. AsAb in each model group rat serum concentrations increased significantly (P<0.05).Then the model was established successfully.2. The sinomenine hydrochloride can improve sperm quality of AsAb immune infertility. The effect was performed in improving sperm density, sperm motility, sperm survival rate, sperm vitality.3. The sinomenine hydrochloride can improve the testis morphology of AsAb immune infertility.
