Objective: Studying the effect of different doses of sinomenine hydrochloride on sperm quality and testicular tissue morphology and sperm acrosome enzyme activity and LDH.Methods:The male sexual maturity (8weeks)48rats randomly divided into sperm and fertility testing before the drug group, blank control group, sinomenine hydrochloride low-dose group (60mg/kg), hydrochloric acid high dose group (240mg/kg),and positive control group. Dosing sinomenine hydrochloride10weeks, observed the different of deformity rate of sperm density、mobility、live sperm rate、sperm acrosome enzyme activity and testis and serum lactate dehydrogenase activity and testicular tissue form in each group rats.RESULTS:1. Viscera coefficient(Testicles, epididymis, seminal vesicle and prostate gland):Compared with blank control group, there are a downward trend on60mg/kg dose hydrochloric acid sinomenine hydrochloride group and240mg/kg dose epididymis coefficient(P<0.05). there are no effect on testes.2. Sperm density:Compared with blank control group, there are no difference in on60mg/kg dose hydrochloric acid sinomenine group and240mg/kg dose hydrochloric acid sinomenine group on sperm density (P>0.05).3. Acrosome enzyme activity:Compared with blank control group, there are no difference in60mg/kg dose hydrochloric acid sinomenine group and240mg/kg dose hydrochloric acid sinomenine group on acrosome enzyme activity (P>0.05).4. Sperm malformation rate:Compared with blank control group, there are no difference in60mg/kg dose hydrochloric acid sinomenine group and240mg/kg dose hydrochloric acid sinomenine group on sperm malformation rate (P>0.05).5. Sperm motility rate、Living sperm rate:Compared with blank control group, there are dropped significantly in positive control group on sperm activity and sperm of male rat live sperm rate(P<0.01);there are no difference in60mg/kg and240mg/kg dose hydrochloric acid sinomenine group in sperm of male rat sperm activity and live sperm rate.6. LDH:Compared with blank control group, there are increased significantly in60mg/kg dose hydrochloric acid sinomenine group on LDH activity of testicular tissue and serum(P<0.01);there are increased in240mg/kg dose hydrochloric acid sinomenine group on LDH activity of testicular tissue and serum(P<0.05).7. There are no difference in each dose group of hydrochloric acid sinomenine on testicular tissue morphology.Conclusion:Each dosing in the male rat (distilled water or corresponding volume) were measured after10weeks. The conclusion is as follows:1. there are no effect significant in60mg/kg and240mg/kg dose of sinomenine hydrochloride on male rats、deformity rate of sperm density、sperm activity and live sperm rate.2.60mg/kg and240mg/kg dose of Sinomenine hydrochloride has no effect on sperm acrosome enzyme activity.3.60mg/kg and240mg/kg dose of Sinomenine hydrochloride has a tendency to rise on lactate dehydrogenase activity.4.50mg/kg methotrexate can reduce significantly the male rat epididymis weights、sperm density、sperm activity, but can not have effect on the sperm acrosome enzyme activity and lactate dehydrogenase activity.5. There are non-toxic effect in60mg/kg and240mg/kg dose of sinomenine on testicular tissue morphology of male rats.
