Objective:This experiment has observed the inhibitory rate of Jianpi Huayu Fang on the transplanted tumor due to colon cell line LoVo in BALB/c mice first, the second step we established an animal model of mice colon cell cancer liver metastasis in BALB/c mice, then observe the effects of Jianpi Huayu Fang on liver metastasis. At last, we has observed Jianpi Huayu Fang’s effects on miRNAs which relating to the colorectal cancer.Methods:1.LoVo cells cultured in vitro were inoculated by using subcutaneous transplantation of BALB/c mice to make subcutaneous tumor nude model mice.30modeling nude mice were randomly divided into negative control group (NS), positive control group (5-FU) and Jianpi Huayu Fang groups (high dosage group, moderate dosage group and low dosage group). The growth, size and weight of nude mice tumor were observed, the inhibitory rate of Jianpi Huayu Fang on subcutaneous tumor in nude mice was analyzed.2.LoVo cells were inoculated into the spleen of BALB/c mouse for making the liver metastasis model.30modeling nude mice were randomly divided into negative control group (NS), positive control group (5-FU) and Jianpi Huayu Fang groups (high dosage group, moderate dosage group and low dosage group). The weight of the tumor of liver metastasis and the rate of liver metastasis were obversed, then analyzed the inhibition of Jianpi Huayu Fang on colon cell cancer liver metastasis.3.Real-time Quantitative PCR was used to detect the expression of miRNAs which relating to the colorectal cancer from subcutaneous tumor and the focus which from the liver metastasis.Result:1.The average tumor volume and the weight of Jianpi Huayu Fang groups (high dosage group and moderate dosage group) were less than those of the negative control group, the difference was statistically significant(P<0.01).2.Jianpi Huayu Fang groups’(high dosage group, moderate dosage group and low dosage group) average weight of the focus which from the liver metastasis were less than those of the negative control group, the difference was statistically significant (P<0.05).3.Jianpi Huayu Fang groups compared with the negative control group, the expression of miRNA-31had been downregulated and miRNA-143been upregulated in subcutaneous tumor. Jianpi Huayu Fang groups compared with the negative control group the expression which from the liver focus of miRNA-31had downregulated. The expression of miRNA-31in liver metastasis tumor had been downregulated.Conclusion:Jianpi Huayu Fang could significantly inhibit the growth of colon cancer and its liver metastasis. The mechanism maybe be relative to regulate the expression of miRNAs.
