| Objectives:To observe the clinical effect of Jianpi Huayu Fang combined with chemotherapy alone and chemotherapy alone in the treatment of advanced colorectal cancer patients,and to evaluate the effects of Jianpi Huayu Fang on advanced colorectal cancer.And the relationship between miRNAs and cytokines,and to explore the possible mechanism of Jianpi Huayu Fang in the treatment of advanced colorectal cancer.Methods:The patients in advanced colorectal cancer were randomly divided into 22 experimental groups and 21 control groups.The patients of experimental group were treated with Jianpi Huayu Fang and Xeloda+ OXA.The control group was treated with Xeloda + OXA regimen alone.After 2 cycles of treatment,Changes of serum cytokines sIL-2R,IL-6 and IL-8 levels in the two groups before and after treatment and the detection of miR-21,miR-155,miR-146a,miR-145in serum by RT-PCR expression changes.Result:(1)Comparison of recent objective efficacy:the two groups after treatment,the difference was not statistically significant(P>0.05).(2)Comparison of KPS score:KPS score was significantly higher in the test group compared with the control group,the difference was statistically significant(P<0.05).(3)Comparison of tumor markers:After treatment,CEA in the experimental group was lower than that in the control group,the difference was significant(P<0.05);CA199 no significant difference(P>0.05).(4)Comparison of miRNAs:The expression of miRNA-21 in both groups was lower than that before treatment,but there was no significant difference between the two groups(P>0.05).The expression of miRNA-146a and miRNA-145 were up-regulated and the amplitude of miRNA-145 was significantly higher in the two groups than in the control group(P<0.05).The expression of miRNA-155 was higher than that of the control group before and after treatment,the difference was statistically significant(P<0.05).(5)Comparison of serum cytokines:The levels of serum IL-6 in the two groups were significantly lower than those before treatment,Compared with the control group,the difference was statistically significant(P<0.05);The levels of serum IL-8 in the two groups were also lower than those before treatment,there was no significant difference between the experimental group and the control group(P>0.05);The levels of serum sIL-2R in the two groups were 7higher than those before treatment,there was no significant difference between the experimental group and the control group(P>0.05).Conclusion:(1)Spleen and blood stasis combined with chemotherapy compared with chemotherapy alone,can significantly improve the physical status of patients with advanced colorectal cancer,there are trends to reduce tumor markers.(2)The mechanism of Spleen Huayu Decoction in advanced colorectal cancer may be through the two-way regulation of miR-21,miR-146a,miR-145 and IL-6 and other cytokines. |
