1. Objective:(1) To find the laboratory-related indexes of idiopathic venousthromboembolism and its value of early diagnosis.(2) Screening for geneticsusceptibility gene of idiopathic venous thromboembolism in Chinese population.2. Methods:(1) We examined laboratory-related indexes(D-dimer, Fibriogen, WhiteBlood Cell, Platelet, ABO blood type) for hospitalized patients of no history ofthrombosis and the first idiopathic venous thromboembolism. Univariate andmultivariate statistical analysis of the correlatio n between these laboratory-relatedindexes and the idiopathic venous thromboembolism.(2) We recorded acquired riskfactors for hospitalized patients of no history of thrombosis and the venousthromboembolism, and applicated-based Sequenom MassARRAY DNA mass arraygene analysis iPLEX Gold snp genotype analysis techniques to test20SNPs whichare related to venous thromboembolism. Univariate and multivariate statisticalanalysis of the correlation between these genetic susceptibility gene and the venousthromboembolism. There contained63patients with idopathic venousthromboembolism in the150patients with venous thromboembolism,and we alsochoosed63patients with age, gender matched in112cases of control group.Univariate and multivariate statistical analysis of the correlation between these geneticsusceptibility gene and the idiopathic venous thromboembolism.3、Results:(1) Through the univariate analysis, D-dimer, Fbg, White Blood Cell andABO blood type were higher in thrombosis group than in the control group (P<0.05),PLT was not difference between thrombosis group and the control group (P>0.05).Through the Logistic regression model analysis, D-dimer and Fbg were the independentrisk factors of the idiopathic venous thromboembolism(P<0.05), the odds ratio were332.79,19.01.the overall partial regression coefficient of95%confidence interval were29.88~3706.49,2.37~152.18. fibriogen>4g/L as the cutoff value,there is significant difference between “thrombosis group” and “the control group”(P=0.00), thesensitivity was89.29%, the specificity was56.63%,positive predictive value was40.98%, negative predictive value was94%; D-dimer>0.5mg/L as the cutoff value,there is significant difference between “thrombosis group” and “the control group”(P<0.05), the sensitivity was92.31%, the specificity was96.61%,positive predictive valuewas96.77%, negative predictive value was91.94%.(2) The acquired risk factors andgenetic susceptibility gene which were significant difference(P<0.2) between between“thrombosis group” and “the control group” used by the univariate analysis wereanalysised by the Logistic.Through the Logistic regression model analysis, age,rs1805087,rs3756008,rs2036914,rs2066865and rs3087505were the independentcorrelation factors of venous thromboembolism in Chinese population(P<0.05), ageOR1.027(95%CI:1.008~1.047). rs1805087-G OR0.281(95%CI:0.105~0.753),rs3756008-T OR0.28(95%CI:0.12~0.653),rs2036914-T OR0.392(95%CI:0.180~0.853),rs2066865-C OR0.381(95%CI:0.163~0.892),rs3087505-T OR5.248(95%CI:1.717~16.043)。Through the univariate and Logistic regression modelanalysis, rs3087505is also the independent correlation factor of idiopathic venousthromboembolism, rs3087505-T OR3.760(95%CI1.009-14.006).4. Conclusions:(1) Laboratory-related indexes(D-dimer,Fbg) were the independent riskfactors of the idiopathic venous thromboembolism. When D-dimer>0.5mg/L, there is avalue of early diagnosis of idiopathic venous thromboembolism. By combination withthe other clinically relevant factors, it may contribute to the early diagnosis of theidiopathic venous thromboembolism.(2) MS、F11、FGG、KLKB1may be the geneticsusceptibility gene of venous thromboembolism in Chinese population, and KLKB1may also be the genetic susceptibility gene of idiopathic venous thromboembolism inChinese population.
