Objectives:Body uncontrolled inflammatory response syndrome after trauma hasbecome the world’s leading cause of death of young adults, bacterial methylationof the CPG-DNA structure is the start of the innate immunity of the body, leadingto uncontrolled sirs key link mtDNA and bacteria on the structure andfunctionsimilar to the activation of NF-κB pathway through TLR-9and produceinflammatory factors, this paper mainly observed mtDNA expression changeswere observed in the severe trauma of the rat peripheral blood mtDNA contentand the relationship of the degree of injury as well as the application ofchloroquine and PDTC on inflammatory markers and importantorgans.methods:Preparation of the first part of the severe trauma rat model: one pairs offemoral shaft fracture rat model;2rat model of hemorrhagic shock model;3pairsof femoral shaft fracture associated with the rat model of hemorrhagic shockmodel. Second part of the rats were divided into four groups: simple pairs offemoral shaft fracture group (n=21)2. Simple hemorrhagic shock group (n=21)3. Pairs of femoral shaft fracture fracture and hemorrhagic shock group (n=21) inthe control group (n=3) rats were divided into four groups: a double-strandedbackbone fracture fracture associated with hemorrhagic shock+chloroquine(10mg/kg) group (n=21) double femoral shaft fractures in the fracture andhemorrhagic shock+chloroquine (30mg/kg) group (n=21) double femoral shaftfracture fracture and hemorrhagic shock+PDTC (30mg/kg) group (n=21)double-stranded backbone two parts of the fracture fracture and hemorrhagicshock+PDTC (120mg/kg) group (n=21) in2hours,4hours,8hours,16hours,24hours,48hours and72hours using the ELISA test assay (ELISA) observationin peripheral blood of TNF-α, IL-10, IL-6expression levels; p65mRNA usingreal-time quantitative PCR (Real-time PCR) was detected in lung tissue NF-κB subunit I-κB mRNA expression of TLR9mRNA; quantitative PCR detection ofperipheral blood mtDNA level; HE staining to observe pathological changes ofthe liver, lung, kidney, intestines. The experimental data were presented as mean±standard error (x±s) represents. All data use SPSS17.0statistical software usingsingle-factor analysis of variance (one way ANOVA).Results:1. Pairs of femoral shaft fractures. Improved combined model of hemorrhagicshock model was a high success rate, low mortality rates, stability andreproducibility can better simulate the clinical high-energy trauma;2.MtDNAcontent in the blood of the rats (2) post-traumatic, blood IL-6, TNF-a levels wereshown a peak showing the trend of gradual decline, and in the subsequent IL-10gradually decreases and then gradually increased trendbut the relative increase inthe concentration of several factors in the three groups of animals showing asignificant difference, the relative increase in hemorrhagic shock group+femoralshaft fracture group was significantly greater than the hemorrhagic shock groupand fracture group;3. The relationship between mitochondrial DNA damage: thedamage is more weight, peripheral blood mitochondrial DNA the higher theconcentration the higher the concentration of inflammatory factors.Lung tissuedamage points higher can be used as early warning indicators of damage;4.serious trauma in rat peripheral blood mitochondrial DNA concentrations andinflammatory markers in lung tissue and p65mRNA, TLR-9mRNA positivecorrelation;5. P65mRNA, I-κB mRNA expression levels in lung tissue negativelycorrelated.;6.Chloroquine and PDTC concentration the higher the lower the levelof expression of inflammatory cytokines, lung tissue damage more points, organdamage, the more light, but the block effect of chloroquine PDTC no significantdifferenceConclusions:The relationship between mitochondrial DNA damage: the damageis more weight, the higher the concentration of peripheral blood mitochondrialDNA, the higher the concentration of inflammatory factors, chloroquine and PDTC could play a protective role in rats with multiple injuries...
