Objective: In this study,by animal experiment, to research restenosis and C-reactiveprotein (CRP) and interleukin1(IL-1) changes after the rabbit carotid arteryatherosclerosis endarterectomy and observe the relationship between the degree ofrestenosis and C-reactive protein combined interleukin-1values. And through theadministration of cilostazol to observe affection of this drug on C-reactive protein andinterleukin-1values and the process of restenosis and to provide a theoretical basis oncarotid restenosis detection and treatment.Materials and methods:60healthy adult New Zealand white rabbits were randomlydivided into three groups: sham operation group, the pure endarterectomyendarterectomy group and endarterectomy plus drug group.All rabbits were given highfat diet for4weeks and then dry medical oxygen was used to injury right carotid arteryintima, and continue to be feeded high-fat diet for8weeks.Except the sham group wasonly dissociated carotid artery in the9th week, the others on the implementation of thecarotid endarterectomy, all continued for high fat diet.The drug group was givenadditionally1mg/kg/d cilostazol orally until killed,and took blood for2ml respectivelyin the ear vein at the2ed,5th,9th,14th,21th day after endarterectomy and then killedimmediately by overdose hardstuff.Take right carotid artery for specimens and werefixed for48hours in10%formaldehyde solution.After been made into paraffin blocks,the specimens were performed pathological examination to observe intima hyperplasiaand calculate intima and media area ratio and detect CRP and IL-1level withenzyme-linked immunosorbent assay.Analyze the data with SPSS19.0software andadopt t test.Results:The two groups showed no obvious endometrial hyperplasia at the2ed day,and CRP had no significant difference in the three groups;IL-1level in endarterectomy plus drug treatment group lower than simple stripping group, and p<0.05showedstatistical significance, but simply stripping group and the sham operation groupshowed no significant difference.Simple endarterectomy group and endarterectomy plusdrug group showed significant intimal hyperplasia from the5th day postoperatively,degree of endometrial exfoliation and drug group lighter than simply stripping group.Tothe14th day proliferation was stable, CRP and IL-1endarterectomy plus drug groupwere lower than stripping group during this process, and p<0.05was statisticallysignificant.CRP and IL-1in the sham group and treatment group endarterectomy had nosignificant difference; the proliferation was substantially similar at the21th day and14th day, but matrix occupy a larger proportion of the components.The hyperplasiaprocess in endarterectomy plus drug group is similar to the simple endarterectomygroup, only slight in degree. CRP and IL-1in both groups has no significant differencein this.Conclusion1.In this study restenosis is mainly occurred within2weeks aftersurgery,the rate of the narrow increased slow down after the2ed week.2.IL-1canreflect the different of the degree of intimal hyperplasia earlier,and the correlationbetween the values of IL-1and the rate of the degree of the rate of intimal hyperplasia isbetter than CRP.3.Cilostazol can reduce the degree of intimal restenosis after carotidendarterectomy.
