Objective To investigate the role of interleukin-17(IL-17) inglomerulosclerosis, and the mechanism of the protective effects of Huai QiHuang (HQH) on adriamycin(ADR)–induced nephropathy in mice.Methods Male BAL b/c mice (8week old) were randomly dividedinto control group, ADR nephropathy group and HQH-treated ADRnephropathy group. ADR nephropathy was induced by a single intravenousinjection of10mg/kg ADR. For the HQH-treated ADR nephropathy group,7days after treatment with10mg/kg adriamycin, the mice wereadministered once-daily oral gavage of4mg/kg HQH for7weeks, whilesaline10ml/kg once a day by gavage in control group and ADR nephropathygroup.24-hour urine was collected at0, second,5thand8thweek after ADRinjection. Six mice were sacrificed at second and8thweek, respectively.Histological changes were observed by light microscopy and transmission electron microscope. Proteinuria was measured by the Bradford proteinassay. Local expression of IL-17in kidney were detected byimmunohistochemistry.Results Compared with control group, mice in ADR nephropathygroup developed overt proteinuria, hypoalbuminemia and impaired renalfunction(all P<0.05). Kidney sections in ADR nephropathy group of the8thweek showed marked focal and even global glomerular sclerosis, associatedwith moderate interstitial expansion and severe infammation. The intrarenalexpression of IL-17was also increased at week8(P<0.05). In contrast, micein HQH-treated ADR nephropathy group had markedly reduced proteinuria,IL-17expression and preserved renal function, with only mild histologicalabnormalities(all P<0.05, compared with ADR nephropathy group).Conclusion HQH is effective in attenuating the renal injury inadriamycin nephropathy. The beneficial effects of HQH are mediated, atleast in part, through reduced local expression of IL-17in kidney. Objective To investigate the ability of interleukin-17(IL-17) toinduce cell growth and extracellular matrix secretion in human mesangialcells,and explore the possible inhibitory effect of Huaier extract.Methods The human mesangial cell line cultured in vitro was dividedinto Normal group, IL-17group, IL-17+Huaier aqueous extract group andHuaier extract group. Cell proliferation and drug cytotoxicity were measuredby MTT test and Trypan blue exclusion test, respectively. The level offibronectin in the culture supernatants was detected by enzyme-linkedimmunosorbent assay (ELISA).Results IL-17could significantly induce cell proliferation andincrease fibronectin production in cultured human mesangial cells. Thiseffect was suppressed with exposure to Huaier extract in a time-anddose-dependent manner.Conclusion The renoprotective effect of Huaier aqueous extract maybe via strongly inhibit the stimulating effect of interleukin-17on humanmesangial cells.
