Objective:To discuss the effect and mechanism on the blood-retinal barrier in the early stage of diabetic SD rats by Intravitreal cyclosporin-A.Methods:Eight-ten weeks old male Sprague-Dawley (SD) rats were used for this study.Diabetes was induced with streptozotocin(STZ). The rats were divided into four groups,with12rats in each group. The four groups were as followed:normal control group (CON); normal control group (CON); diabetic control group (DM); diabetic with CsA intravitreal injection group (DM+CsA); diabetic with DMSO intravitreal injection group (DM+DMSO). CsA and DMSO were injected into the vitreous of eyes4weeks after induction of diabetes.Retinas were harvested48h following treatment. BRB permeability was quantitated by measuring extravasated endogenous albumin, ICAM-1and VEGF levels in retinas were studied by Western blot and enzyme-linked immunosorbentassay (ELISA) respectively.Results:Compared with diabetic retinas,Diabetes-induced retinal albumin extravasation were significantly reduced in CsA-treated diabetic retinas(P<0.05), the ICAM-1and VEGF expression in the retinas were significantly reduced(P<0.05).Conclusion:CsA may stabilize the BRB in association with down-regulation of ICAM-1and VEGF expression in retinas in the early stages of diabetes.
