Objective: To study effects of different concentrations of quercet in human retinal capillary endothelial cells(HRCECs)proliferation and the expression of vascular endothelial growth factor(VEGF)and ICAM-1in high glucose environment.Methods:1.Cultured human retinal capillary endothelial cells(HRCECs)invitro,and select the well growth of 3-4 generation.2.At 24?48?72hours cultured,Detecting the multiplication on different concentrations of quercetin of HRCECs under high glucose environment by MTT.3.Then Western Blot is used to detect the expression of VEGF and ICAM-1 in different concentrations of quercetin in HRCECs under high glucose? Results: 1?MTT assay results has showed that:? after HRCECs culture 24,48,72 hours later,at the same time point,the absorbance value of was higher than that of the low glucose control group,The difference was statistically significant(P<0.05),and in the high glucose group,the absorbance value increased with the extension of time;? Different concentrations of quercetin used to separate culture HRCECs24,48,72 hours in high glucose environment,at the same time point,Compared with the control group,The absorbance value of HRCECs decreased,The difference was statistically significant(P<0.05),and with the increase of the absorbance value decreased gradually;The absorbance values in different concentrations of quercetin group were compared at the same time point,The difference was statistically significant(P<0.05),And with the increase of the absorbance value decreased gradually;the absorbance values in the same concentration of quercetin group were compared with each other at different time points,The difference was statistically significant(P<0.05),?2?Effects of quercetin on the expression of VEGF and ICAM-1 in HRCECs under the high glucose ?In the low sugar control group,HRCECs after 24,48,72 hours of culture,VEGF and ICAM-1 were expressed in a small amount,no apparent time dependence,the difference was not statistically significant(P>0.05)?Compared with low sugar control group,the expression of VEGF and ICAM-1 was up-regulated in high glucose environment,And with the extension of time,the higher expression,the difference was statistically significant(P<0.05),and with the extension of time,the absorbance value decreased gradually? ?Effects of different concentrations of quercetin on HRCECs 24 h,48h,72 h after high temperature,at the same time point,Compared with the high glucose control group,The expression levels of VEGF and ICAM-1 were decreased in different concentrations of Quercetin,The difference was statistically significant(P<0.05),And with the increase of concentration,the expression of both decreased;The expression of VEGF and ICAM-1 in different concentrations of quercetin group were compared with each other at the same time point,the difference was statistically significant(P<0.05),And with the increase of concentration,the expression of both declined;the expression of VEGF and ICAM-1 in the same concentration of quercetin group were compared with each other at different time points,the difference was statistically significant(P<0.05),And with the increase of time,the expression of both declined? Conclusion:1.Quercetin could inhibit the proliferation of HRCECs cultured in vitro under high glucose concentration,and in a time-and concentration dependent;2.Quercetin could decrease the expression of VEGF and ICAM-1 in the growth of HRCECs under high glucose in vitro,and in a time-and concentration dependent;3.The inhibition mechanism of quercetin on the growth of HRECs in vitro may be achieved by down regulating the expression of VEGF and ICAM-1 in HRCECs.
