Objective To investigate the effect of Oleic Acid(OA) on the expression ofPGC-1α mRNA and protein in mouse hepa1-6cells and to unravel the signalparthway of the process.Methods In this study, mouse hepa1-6cells were treated by differentconcentrations of OA at various time,and then PGC-1αmRNA levels were detectedby RT-PCR.The same factors treated for12h,the apoptosis rate was examined by flowcytometer.Treated cells by OA and SB203580(inhibitor of p38MAPK),and theProtein expression of PGC-1αwas assessed by Western Blot.Results1、Mouse hepa1-6cells were treated by various concentrations of OA for2h respectively,compared with control group,the expression of PGC-1a mRNA wasincreased at0.2mmol/L OA concentration (P <0.05), but it showed no significantdifferences at the OA concentrations of0.1mmol/L group、0.5mmol/L group and1.0mmol/L group.2、Mouse hepa1-6cells were treated by various concentrations of OA for12h respectively,compared with control group,the expression of PGC-1a mRNA wasincreased at0.2mmol/L and0.5mmol/L OA concentration (P <0.01), but it showedno significant differences at the OA concentrations of0.1mmol/L group and1.0mmol/L group.3、Mouse hepa1-6cells were treated by various concentrations of OA for24hrespectively,compared with control group,the expression of PGC-1a mRNA wasincreased at0.2mmol/L and0.5mmol/L OA concentration (P <0.05), but it showedno significant differences at the OA concentrations of0.1mmol/L group and1.0mmol/L group.4、Mouse hepa1-6cells were treated by0.2mmol/L OA and20μmol/LSB203580for24h,the expression of PGC-1a protein was increased at the0.2mmol/L OA group (P<0.01versus control).However, the expression of PGC-1a protein wassignificantly decreased at the OA with SB203580group(P <0.01versus OA group)5、Mouse hepa1-6cells were treated by various concentrations of OA for12h,the rate of apoptosis was increased in a dose-dependent manner with OA(P<0.05versus control).Conlusions OA could promote the transcription of PGC-1α mRNA and increasethe protein expression of PGC-1αin Mouse hepa1-6cells,and the effect of OA on thePGC-1α gene expression might be related to p38MAPK signaling pathways.The rateof cellular apoptosis was increased in a dose-dependent manner with OA.
