Objective To observe the concentrations of SOD,MDA.NO.ET-1and expression of apoptosis indexes of skeletal muscle.and investigate the protective mechanism of Shenmai injection on limb ischemia-reperfusion injury in rats.Methods24SD rats were randomly divided into two groups (n=8):Ischemic reperfusion+NS group (I/R group). Ischemic reperfusion+Shenmai injection group(SM group). Ischemic reperfusion models were made by the femoral vessels ligation. Ischemia-reperfusion injury was not induced in the sham-surgery group (S group)。The concentrations of the SOD, MDA. NO and ET-1in the blood samples were determined.The Immunohistochemistry (IHC) method was used to detect the expression of Bcl-2and Fas in gastrocnemius tissues.Results①As compared with the sham-surgery group(S group),the SOD activity in the ischemic reperfusion+NS group(I/R group) was significantly less (P<0.01), the MDA content was significantly greater (P<0.01).The model was successful and involved in the limb ischemia-reperfusion injury model.And compared with the ischemic reperfusion+NS group(I/R group), the SOD activity in the ischemic reperfusion+Shenmai injection group(SM group) was significantly greater (P<0.01).the MDA content was significantly less (P<0.01).②ompared with the ischemic reperfusion+NS group(I/R group), the level of ET-1in plasma in the ischemic reperfusion+Shenmai injection group(SM group) was decreased (P<0.01), NO and the percentage of NO/ET-1were increased(P<0.01).③As compared with the sham-surgery group(S group), the Fas and Bcl-2 in gastrocnemius tissues were significantly greater (P<0.01) in the ischemic reperfusion+NS group(I/R group). And as compared with the ischemic reperfusion+NS group(I/R group), the Fas in gastrocnemius tissues was significantly less (P<0.05). but Bc1-2was significantly greater (P<0.05) in the ischemic reperfusion+Shenmai injection group(SM group).Conclusion①The levels of oxygen free radicals were increased by following the ischemia reperfusion injury.②The disturbance of nitric oxide/endothelin-1(NO/ET-1) was induced by the ischemia-reperfusion injury.③Shenmai injection can resist lipid peroxidation of free radicals.④Shenmai injection can protect the endothelial cells.⑤Shenmai injection can inhibitskeletal muscle cell apoptosis. protect skeletal muscle cells after ischemia-reperfusion injury.⑥The application of Shenmai injection will be an effective solution for the ischemia-reperfusion injury in clinic.
