ObjectivesTo further explore the role of HBV in hepatocellular carcinoma (HCC) and its possible carcinogenic mechanism.Methods1.The selection and collection of specimen of the cases.2.Experimental grouping. Case specimens were divided into three groups, respectively for liver cancer, liver tissue by Carcinoma and normal liver tissue group.3.Immunohistochemical S-P method to detect the expression of the AFP and HBsAg in the tissue samples. Respectively with immunohistochemical method to detect the expression of the AFP and HBsAg in organization paraffin of the tissue samples and record the results under a microscope.4. The expression of HBV-DNA on the paraffin section of the HBsAg positive HCC tissue and liver tissue by cancer and the normal liver tissue was detected respectively by In situ hybridization technology (RT-PCR);the results was observed and recorded under the microscope.5. The expression of AFrl on the paraffin section of the HBsAg positive HCC tissues and liver tissue by cancer and the normal liver tissue was detected respectively by Western blot method;the results was observed and recorded under the microscope.Results1. The expression of HBsAg and AFP on all normal liver tissue were negative. The expression of HBsAg on the cancer and liver tissue by HCC is respectively70%,90%(P<0.05). The expression of AFP on the cancer and liver tissue by HCC is respectively were87.5%,82.5%(P>0.05). The AFP express on positive HBsAg HCC and the liver tissue by cancer was significantly higher than the negative HBsAg (P<0.05).2. The positive signal on HCC and liver tissue by HCC of HBsAg positive is more and stronger than normal liver tissues. HBV-DNA positive physical form have cytoplasmic type, karyotype and nuclear plasma type, can be alone or mix exist.3. AFrl only expressied on the HCC (7.1%,2/28) and liver cell by cancer tissues (0%,0/28), But it do not express on normal liver group. The HCC tissue has a very significant difference with normal liver tissue (P<0.01).ConclusionsHBV infection may induce liver cells producing AFP to participate in HCC disease through gene integration, the specific molecular biological mechanisms need further research.
