Objective To investigate the mechanism of HBVDNA integration and the nosogenesis of hepatocellular carcinoma by detecting the expression of HBV P gene and the integration of HBV P , X gene in PBMCs of HCC patients. Method Selected 40 HCC patients with HBsAg(+) in this study. 5 HCC patients with HBsAg(-) and 5 cases of healthy volunteers were for control groups. Extracted RNA and DNA from isolated PBMCs of these patients' peripheral blood samples separately, then reversed total RNA to cDNA. Primers were designed for PCR to determine the integration of HBV P, X gene in PBMCs. RT-PCR was used to determine the expression of HBV P Gene .Results X gene integration was 79.5%(31/40). P gene integration was 15%(6/40). The integration was significantly different between X and P gene(P<0.01). The expression of HBV P gene was 10% (4/40) ,which was lower than 33. 3% (12/36) in the chronic hepatitis patients (P<0. 05). Further study indicated that there was no significant difference of HBV DNA integration and expression in HCC three different stages. So was HBV DNA positive and negative cases(P>0.05). No expression of HBV P gene and no integration of X gene or P gene was found in PBMCs in the control and healthy volunteers groups.Conclusion The integration of X gene was significantly higher than P gene in PBMCs of HCC patients . The replication of HBV was inactivity in this stage .The integration and expression of X gene played a major role in most HBV-induced HCC.
