Objective:Through the establishment of SANFH rabbit model, it is observed, in the treatment process of animal models, that what’s the affection of bone erosion ning No.1and No.2 on the expression of VEGF, and what are the situations of the happening of repair the femoral head and regenerations in each periods. It is to explore the bone erosion ning on the mechanism of action to glucocorticoid-induced avascular necrosis.And reliable basis can be provided for the clinical application of bone erosion ning.Methods:1.Effect of bone erosion ning on the expression of VEGF in SANFH rabbits1.1 60 adult and healthy adult New Zealand are divided into 4 groups in random:6rabbits in normal group; 18 rabbits in bone erosion ning group; 18 rabbits in blank control group(normal saline group); 18 rabbits in positive control group(Jian-Gu-Sheng-Wan). What’s more, all have their numbers for identification. As a comparison standard, the normal group feed as usual in a single cage. Rest of them are used in the model. After this, the control group will have lavages on a regular basis. The positive control group will also have lavages on a regular basis after adopting distilled water dissolving. According to TG(in normal 1.00±0.72mmol/L), bone erosion ning group takes 1.00mmol/L as a standard. The rabbits with a higher number than this are dialectically thought as blood stasis models, which will drench bone erosion ning No.1later. Rest of them is dialectically thought as kidney deficiency models, which will drench bone erosion ning No.2. All of them respectively put to death in 4 weeks, 8weeks and 12 weeks. Bilateral femoral head specimens are taken to paraffin-embedded.1.2 Taking tissue sections of the caput femurs of paraffin-embedded, and taking SABC method to partly detect the expression of VEGF.2. Bone erosion ning’s influence on SANFH rabbits about model histopathologic change2.1 Experimental steps as the above.2.2 By HE staining, the microscopic observation of femoral head organization form, and calculation of the empty pit of bone.Results:1.Measuring the expression of VEGFAs rabbits are adapted to the fourth week, the VEGF intensity of the blank control group is slightly below that of the normal group(P<0.05). Whereas the intensity of Jian-Gu-Sheng group and bone erosion ning group are remarkably decreased by comparison with that of blank control group(P<0.05). In the eighth week, intensity of the blank control group is apparently higher than the rest three groups. And there is a statistical discrepancy(P<0.05). In the twelfth week, the expressions of VEGF of Jian-Gu-Sheng group and bone erosion ning group are significantly high(P<0.05).However, there is not any statistical discrepancy among the two groups and the normal group( P>0.05). The intensity of bone erosion ning group is lower than the Jian-Gu-Sheng group(P<0.05).2.Observation of HE dyeingAfter 4 weeks, Model groups show that trabecular trabecular bone thinning,osteoblast decreases, fat cells increased,Jian-Gu-Sheng group and bone erosion ning group show that trabeculae getting thinnner,less trabecular number,bone marrow filled with hypertrophy cells;After 8 weeks, Jian-Gu-Sheng group and bone erosion ning group show:the number of osteoblast cells and trabecular trabecular bone is higher than the blank control group,and Osteoclast decreased;After 12 weeks, Jian-Gu-Sheng group and bone erosion ning group show:trabecular structure was complete basicly.Conclusion1.The rise of the expression of VEGF enables to induce partial restoration of caput femoris.2.Bone erosion ning can enhance the expression of VEGF.3.Through improving the partial’s blood supply, bone erosion ning make a contribution to the prophylaxis and treatment of SANFH.
