OBJECTIVETo investigate the effect of SRP and the mechanism,we determined the behaviorsand trace elements,the ratio of mean Zn/Cu and monoamine neurotransmitters5-HT inthe blood serum of the rats with chronic unpredictable mild stress (CUMS) induceddepression.METHODSThirty six SD rats were divided into six groups: normal, control, depression model, fluoxetine, SRP (100,200,400mg/kg) groups. Rat models were exposed to alternatestressors for21days to induce depression. Behavioral indexes were measured, fourkinds of trace elements, the ratio of mean Zn/Cu and5-HT content in blood serum ofnormal control, depression model, fluoxetine, SRP groups were measured by the atomicabsorption spectrophotometer and fluorescence chromatogram.RESULTS1.The behavioral indexes showed an obvious depressive state in rats of model groupas compared with normal control rats.2.Compared with normal control group, content of Zn, Mn, Mg in the serum of modelgroup decreased obviously, but that of Cu increased (P<0.01); and the ratio ofZn/Cu decreased obviously (P<0.01).The results of SRP groups are same withfluoxetine group, content in the serum of Zn, Mn, Mg and the ratio of mean Zn/Cuincreased obviously, and that of Cu decreased(P<0.05, P<0.01).There issignificantly positive correlation in the level of serum Zn and the ratio of meanZn/Cu in each groups(r=0.877, P<0.01);and there is negative correlations betweenserum Cu and the ratio of mean Zn/Cu in each groups(r=-0.948, P<0.01).There issignificantly positive correlation in the level of serum Mg and the ratio of meanZn/Cu in each groups(r=0.914, P<0.01);the relation with serum Mn and the ratio ofmean Zn/Cu in each groups is also significantly positively correlated(r=0.911,P<0.01).3.Compared with normal control group, the content of5-HT in the serum of modelgroup decreased obviously (P<0.05). Compared with model group, the content of5-HT in the serum of SRP(100,200,400mg/kg) groups increased obviously(P<0.05). There is significantly positive correlation in the level of serum5-HT andZn in each groups(r=0.927, P<0.01);and there is negative correlations between serum5-HT and Cu in each groups(r=-0.894, P<0.01);there is significantlypositive correlation in the level of serum5-HT and Mg in each groups(r=0.786,P<0.01);there is significantly positive correlation in the level of serum5-HT andMn in each groups(r=0.898, P<0.01);the relation with serum5-HT and the ratio ofmean Zn/Cu in each groups is also positively correlated (r=0.768, P<0.01).CONCLUSIONS1. SRP could improve behaviors of rats with the chronic stress depression model,which has anti-depressive effect.2. SRP could heighten the level of Zn, Mn, Mg, reduce the level of Cu, andheighten the ratio of Zn/Cu in the serum, that maybe the possible mechanism ofthe anti-depression of SRP.3. SRP could heighten the level of monoamine neurotransmitters5-HT in theserum, that maybe the possible mechanism of the anti-depression of SRP.
