Objective To explore the method and therapeutic effect of neural stem cellstransplantation for spinal cord injury.Methods The neural stem cells were isolated from the spinal cord, purified andcultured. After three generation, this neural stem cells was concentrated and stored in4℃refrigeratory. Spinal cord injury was made in all these rats at T10with modifiedAllen’s method.60animal spinal cord injury model was divided into three grouprandomly(n=20). One spinal cord injury group received transplantion of single neuralstem cell suspensions. Another group received transplants of neurosphere cellsuspensions.3、7、14、21、28days later, this nouse was killed and spinal core wasextracted. On the same day locomotor abilities were assessed using the Basso, Beattieand Bresnahan (BBB) rating scale. Subsequently using Immunofluorescence to markneuron and count the neuron. The same time calculate the neural stem cells’differentiation rate and analysis using statistical knowledges.Result After3days of the transplant team, Basso-Beattie-Bresnahan score of SD ratswith spinal cord injury showed not significant improvement and was graduallyincreasing with time. Until7days of transplantation hindlimb motor function improvedslightly, but there was no significant difference. meanwhile compared to two controlgroups, the number of MAP-2positive cells of experimental group has no statisticaldifference after three and seven days(P>0.05). After14days、21days、28days, Basso-Beattie-Bresnahan score of experimental group was (14.8±1.5)、(16±0.8)、(17.8±2.4). The score was better than the other two control groups by statisticalanalysis(P<0.05), and there was statistical significance by comparing the control group2to the control group1. The number of MAP-2positive cells of14days、21days、28days were(17.0±1.0)、(18.8±1.5)、(19.4±3.6),and more prevalent in the transplantgroups than the two controls after7days by statistical analysis(P<0.05)。Conclusions Transplants of neurophere suspensions can promote neural stem cells todifferentiate into neuron and are more conducive to functional recovery after spinal cordinjury.
