Objective:To explore the function of NF-κBp65(nuclear factor κB p65)and IκBα(inhibitor of nuclear factor κB α) in the development and metastasis of gestational trophoblastic tumor by analyzing the expressin of NF-KBp65and IκBα in normal pregnancy villi and gestational trophoblastic diseases,To research the relationship between NF-κBp65and IκBα with age and clinical stages factors of gestational trophoblastic tumors,and explore the role of NF-KBp65and IκBα in gestational trophoblastic tumor occurrence and development.Methods:The expression of NF-κBp65and IκBα were detected by immunohistochemistry(S-P) in normal pregnancy villi(20cases), hydatidiform moles(30Cases), invasive hydatidform moles(13Cases) and chorionic carcinomas(15Cases).Results:1. The positive rate of NF-κBp65protein in normal pregnancy villi,hydatidiform moles,invasive hydatidiform moles and chorionic carcinomas were30.0%(6/20),40.0%(12/30).71.9%(10/13)、73.3%(11/15).The difference between normal pregnancy villi group and invasive hydatidiform moles group,normal pregnancy villi group and chorionic carcinoma goup,hydatidiform moles group and invasive hydatidiform moles group,hydatidiform moles group and chorionic carcinomas goup were statistically significant(P=0.013, P=0.018, P=0.026, P=0.035,P<0.05).The expression of NF-KBp65protein was correlated to clinical stage, while there was no close relation to ages.2. The positive rate of IκBa protein in normal pregnancy villi,hydatidiform moles,invasive hydatidiform moles and chorionic carcinomas were95.0%(19/20),86.7%(26/30),46.2%(6/13),33.3%(5/15).The difference between normal pregnancy villi group and invasive hydatidiform moles group,normal pregnancy villi group and chorionic carcinomas goup,hydatidiform moles group and invasive hydatidiform moles group,and hydatidiform moles group and chorionic carcinomas goup were statistically significant(P=0.003, P=0.000, P=0.009, P=0.001,P<0.05).The expression of IκBα protein was correlated to clinical stage, while there was no close relation to ages.3. By Spearman correlation analysis showed that, there was negative relationship between the protein expression of NF-κBp65and IκBα in GTN (r=-0.403, P=0.034, P<0.05).Conclusion:1. The expression of NF-KBp65in gestational trophoblastic tumor was higher than in normal pregnancy villi and hydatidiform moles, and with the clinical stage of progress, its expression was higher,which indicated that NF-KBp65may be related to the development of gestational trophoblastic tumor, and may be involved in trophoblastic tumor invasion and metastasis.2.The expression of IκBα in gestational trophoblastic tumor was lower than in normal pregnancy villi and hydatidiform moles,and with the clinical stage of progress,its expression was lower,which indicated possibly the reduction or absence of IκBα may be related to the malignant transformation of trophoblasts.3. NF-KBp65, IκBα both in gestational trophoblastic tumor tissues was negatively correlated.The reduction or absence of IκBα may increase NF-KBp65by promoting the development of gestational trophoblastic tumor, and invasion and metastasis.
