PurposesThrough animal experiments, the effect and feature of the platelet-rich gel without activators guiding the bone regeneration in the repair of bone defect can be evaluated. In order to provide a reference for clinic, the osteogenesis effects of the new method should be compared with the platelet-rich gel with activators. Whether the activator caused coagulant disorders is necessary to detected.MethodsAll of the36rabbits for research are penetrated bone defect, and all of them are filled with the platelet-rich gel. Examining the hematoxylin-eosin staining (HE staining), Masson trichrome staining and X-ray, the status of bon defects in2weeks,4weeks,8weeks, and12weeks will be Analyzed. The effect of bone defect repair of the new method should be compared with the platelet-rich gel with activators. For analyzing the effect of the activators to animals’coagulation function, the experimental group and the control group should be compared.Results1. After surgery, the animal for research has no complication, for example, wound infection, and dehiscence.2. The X-ray image of the bone tissue shows the all of the methods have osteogenesis effects. The periosteum is higher than control group (P≤0.05)3. The sclerous tissues slice of bone samples show all of the two methods can make the bone regeneration, and repair the bone defects completely. The effect is higher than control group (P≤0.05).4. There has no obvious difference about the two platelet-rich gels in osteogenesis effects by statistical analysis (P>0.05).5. The coagulation indexes of the platelet-rich gel with activators at each time point have a slight change. The vertical comparison has less statistical significance, but the lateral comparison shows that the coagulation indexes of the platelet-rich gels with activators in2weeks,4weeks,8weeks, and12weeks are different, and the difference has statistical significance.ConclusionsThe platelet-rich gels that prepared by the two methods are all can guide the bone regeneration, and repair the bone defects completely. The coagulopathy by bovine thrombin for rabbits has not been found, but with the time change, the coagulation indexes have a slight change. Whether the bovine thrombin can cause coagulation disorders or not remains to be determined.
