Clinical epidemiology survey:fixed crowns and bridges, abutments prone to gum inflammation, prosthesis edge of the seal is one of the main gingival inflammation. Not close together, crowns and bonding after the binder did not go to the net so that the binder after the bonding of fixed prostheses in the gingival crevicular fluid exposure.Neutrophils in the gingival crevicular fluid is the first line of defense of the periodontal innate immune from the peripheral blood circulation. With sterilization and release of inflammatory factors such as the role of neutrophils. Some associated with neutrophil number to reduce or functional defects of systemic disease patients frequently suffer from periodontitis. Therefore, neutrophils are essential to the health of the periodontal tissue.Studies have found that neutrophils can effectively kill and remove bacteria, but when the face of a too large and can not be swallowed material, known as "frustrated phagocytosis"."Frustrated" neutrophil activity will be weakened, membrane permeability increased, and increased secretion of inflammatory factors. The combination of the above point of view, to consider whether the binder and neutrophil contact, it will reduce its vitality, increase cell membrane permeability, increased secretion of inflammatory factors, and the fixed prosthesis prone to gum inflammation. In this study, direct contact of neutrophils with different binders training, to detect the survival rate of apoptosis rate and necrosis rate, secretion of IL-8and ROS, and then explore the exposure of the binder is related to gingival inflammation and the experimental results of comparing different binder, provide a theoretical basis for the clinical selection ofTest I Induction of The early apoptosis, necrosis rate and survival rate by three different prosthodontics cements on human Polymorph nuclear neutrophils. Objective:To evaluate the capacity of effects of three cements on The early apoptosis, necrosis rate and survival rate of polymorph nuclear neutrophils. Methods:Glass ionomer cement, zinc polycarboxylate cement, light-cured resin cement line the bottom of24-well plates. Polymorph nuclear neutrophils (PMNs) were isolated from blood collected from healthy volunteer donors. Neutrophils were incubated in wells for4hours. Flow cytometry test cell viabilityResults:the early apoptosis and necrosis rate of the zinc polycarboxylate cement group is highest, the survival rate is the lowest; the early apoptosis and necrosis rate of glass ionomer group is lowest, survival rate is the highest; the early apoptosis rate, the necrosis rate, the survival rate of the experimental groups were higher than the Negative group, which had significant difference to the Negative group.(P<0.05).Conclusions:Glass ionomer displays relative excellent antibacterial activities and lower inflammatory activation. Test II Induction of IL-8by three different prosthodontics cements on human Polymorph nuclear neutrophils. Objective:To evaluate the capacity of effects of three cements on concentration of interleukin-8(IL-8) of polymorph nuclear neutrophils. Methods:Glass ionomer cement, zinc polycarboxylate cement, light-cured resin cement line the bottom of24-well plates. Polymorph nuclear neutrophils (PMNs) were isolated from blood collected from healthy volunteer donors. Neutrophils were incubated in wells for4hours.. Concentration of interleukin-8(IL-8)in supernatant fluid was detected by ELISA.Results:The concentration of il-8of the cements is significantly higher than the control group. The glass ionomer cement group is the highest, and the zinc polycarboxylate cement group lowest. Conclusions:The glass ionomer displays characteristics of lower inflammatory activation. Test III Induction of ROS by three different prosthodontics cements on human Polymorph nuclear neutrophils. Objective:To evaluate the capacity of effects of three cements on concentration of reactive oxygen species (ROS) of polymorph nuclear neutrophils. Flow cytometry test cell viability and reactive oxygen species (ROS) in PMNs.Results:The ROS level of neutrophils demonstrates a significantly increase. Conclusions:Glass ionomer displays relative excellent antibacterial activities...
