Research Of You Guiwan On Th17/Treg Gene Expression Level In Mice With Immune Premature Ovarian Failure(POF)Hunan University Of Traditional Chinese Medicine

Objective:To observe the effect of Youguiwan on Th17/Treg gene expression level in mice with immune premature ovarian failure (POF).Methods:In January age BALB/C female mice as experimental objects. Model of POF was established by multiple point injection in pairs of soles and obdaminal subcutaneous with pZP3.After injection,mice was devided to Normal group, Model group, western medicine group, Chinese and western medicine group and Yougui group. Normal group and Model group was treated with distilled water,and other groups were respectively treated with Youguiwan,sex hormone sequential and union of Chinese and western medicine.The mice were in January treatment after been acrificed and taking ovarian,and then we observe the morphological changes of it’s ovarian tissue under the light microscope with HE staining.ELISA method for the detection of serum E2and FSH levels; RT-PCR method for the detection of ovarian tissue in Th17and Treg gene expression levels; SPSS18.0software package was used for statistical analysis.Results:The model group compared with normal group in serum E2content and tissue Treg geng expression level was significantly reduced (P<0.01),but in FSH content,Th17gene expression and Th17/Treg level was notably increased(p＜O.O1); The indictors of Youguiwan group and normal group had no statistica significance;Youguiwan group compared with model group,that except Treg gen expression level were not statistically significant, more than all have significar difference (P<0.01); Western medicine group or Chinese and western medicine grou compared with model group in Th17gene expression level and Thl7/Treg level i increased dramatically (P<0.01), and is a notable decrease compared with norma group(P<0.05); There is no obvious difference between western medicine group Chinese and western medicine group and Yougui group with normal group and mod(group in Treg gene expression level (P>0.05).Conclusions:1.Ovarian tissue Th17gene obviously up-regulated, Treg gen expression clear cut and Th17/Treg express imbalance, which have a clos relationship between autoimmune premature ovarian failure;2.Youguiwan can adjus the autoimmune POF model of mice endocrine (FSH and E2) and the Th17/Tre gene expression balance;3.Youguiwan on Th17/Treg balance adjustment may be th balance regulating ways on kidney yin and yang.
