Objective To explore the effect of Buyang Huanwu decoction(BYHWD) on the neurological function, the area of infarction, estrogen-2(E2) and estrogen receptor(ER) level in middle cerebral artery occlusion (MCAO) ovariectomized-rats.Methods36female SD rats were randomly divided into following6groups:normal group, MCAO group, sham operation group, model group (OVX+MCAO group), BYHWD group, estradiol group. The rats were operated ovariectomy first. The ovariectomized rats model were evaluated by vaginal secretion smear test in10days later. And then, they were divided into groups by the weight and accepted MCAO operation. After MCAO operation, they were fed BYHWD for7days by using stomach tube. We observed the rats on the neurological function at2hours after MCAO and7days medication. The area of infarction was calculated after TTC detection. E2and ER level in serum were determined by enzyme linked immunosorbent assay.Results1.There were obvious neurological defects on every group of MCAO ovariectomized-rats. The rats’neurological score reduced with the survival time. It suggests that the rats own have self-repair capabilities; there is no diference between MCAO and model group; compared with model group, there was statistically significant on BYHWD for decreasing the neurological score (P<0.05).2.The MCAO ovariectomized-rats brains were stained by TTC, their left subcortical presented white infarct area. It suggests the infarcted was different. After the7days medication, there is no diference between MCAO and model group on the infarct area of the rats; compared with model group, there was statistically significant on BYHWD group (P<0.05)3.After the7days medication, the E2level in rats serum were detected the by doing Elisa test. Campared with model group, there was statistically significant on BYHWD for improving the E2level (P<0.05); there is no diferece between BYHWD group and normol group.4.After the7days medication, the rats ER level were detected by using Elisa test. BYHWD can significantly increase the expression of ER in MCAO ovariectomized-rats serum. Campared with model group, this increase was statistically significant (P<0.05); the ER level of BYHWD group is similar to normol group.Conclusion1. BYHWD can reduce infarct size and promote the recovery of neurological function on MCAO ovariectomized-rats.2. BYHWD can raise the levels of E2and strengthen the expression of ER in the serum of MCAO ovariectomized rats.
