This project was supported by national natural science foundation (No.30772785, NO.30973939),"Eleven-Five" national projects of technological supporting (No.2006BAI09B06-10).Pinellia ternate(Thunb.)Breit. and Pinellia pedatisecta Schott are all belonged to Araceae family. The property and flavor of the four Araceae plants is all documented to be’acrid, warm and toxic’. These crude drugs have intensive acridity and toxicity, causing swelling of the lips, mouth and throat, aphonia, salivation, convulsion, and even death from suffocation. The needle-like raphides of calcium oxalate were proved to be their common poisonous components, named as toxic raphides. The raphides caused severe conjunctival edema in rabbit, caused toe swelling of rat. Toxic raphide were a complex compsed by mainly calcium oxalate crystals and protein. The agglutinin on the toxic raphide can significantly induce rat peritoneal neutrophil migration to peritoneal and rat paw edema. The pro-inflammtation effect of PTA could work only when the raphides pierce into the organization, and PTA enhanced the inflammation induced by raphides.So clinical medication is that they are all processed with Potash Alum through duplicating method. The alum (Mainly containing aluminum potassium sulfate dodecahydrate)main function is to reduce its irritant toxicity. Potash Alum cause the decomposition of calcium oxalate, and the rigid support of crystal is damaged. Speculated thatmay be associated with the combination of C2O42-and Al3+.This article studies on the common mechanism of detoxification processing with Potash Alum in Pinellia ternate(Thunb.)Breit. and Pinellia pedatisecta Schott.Main contents and findings were the following:1Pro-inflammatory effect of Pinellia ternata agglutinin and Pinellia pedatisecta agglutininTo further study the pro-inflammatory effect of Pinellia ternata agglutinin (PTA) and Pinellia pedatisecta agglutinin (PAA), the model of Rats peritonitis was used to study the dose-toxicity and time-toxicity relationship of the effect by detecting the releases of inflammatory mediators PGE2in the exudates; The model of Draize rabbit eye test was applied to determine the correlation of PTA,PAA and toxicity of raphides by pathological examination. PTA and PAA enhanced the content of PGE2and protein in rats peritoneal cavities concentration dependently; With PTA and PAA concentration increased, they enhanced the inflammation induced by raphides to rabbit eyes, but they alone had no toxicity response. So PTA and PAA had obvious effect of pro-inflammation. The toxic mechanism of Pinellia ternata was PTA and PAA induced inflammation only when the raphides pierce into the organization.2Influence of Pinellia ternata(Thunb.)Breit. and Pinellia pedatisecta Schott irritating induced by alumbreThe models of Draize rabbit eye test and mouse peritonitis was used to study the Influence of Pinellia ternata(Thunb.)Breit. and Pinellia pedatisecta Schott irritating induced by alumbre. The result showed that8%Alum solution can reduce the irritating of toxic raphides to rabbit eye. And agglutinin which was processed by alumbre can reduce the releases of inflammatory mediators PGE2and protein.8%Alum solution can reduce the irritating toxicity of toxic raphides and agglutinin.3Common mechanism of detoxification processing of Pinellia ternate(Thunb.)Breit. and Pinellia pedatisecta SchottScanning electron microscope was used to stuy the changes in the needle crystal, SDS-PAGE was used to stuy the changes in protein,27Al-NMR was used to study the combination of C2O42-and Al3+. The result showed that Al3+in alaum water solution have the combination ability with C2O42-which comes from calcium oxalate raphides, which destroyed the raphides structure and crystal form; SDS-PAGE was used to study the influence of the agglutinin induced by the alumen. Agglutinin dissolved in8%Alum solution and was degradation.Through the researches above, Pinellia ternate(Thunb.)Breit. and Pinellia pedatisecta Schott have similar toxic components:toxic raphides and agglutinin. The agglutinin had obvious effect of pro-inflammation. They are all processed with Potash Alum through duplicating method. Al3+in alaum water solution have the combination ability with C2O42-which comes from calcium oxalate raphides, which destroyed the raphides structure and crystal form; Agglutinin dissolved in8%Alum solution and was degradation.
